【摘要】：目的研究沙格列汀片在精神分裂癥合并2型糖尿病患者中多次給藥后的藥動(dòng)藥效學(xué)。方法 10例精神分裂癥合并2型糖尿病男性住院患者口服沙格列汀片5 mq qd 7 d,在第7天采集12 h內(nèi)動(dòng)態(tài)血標(biāo)本,采用HPLC-MS/MS測(cè)定血漿中沙格列汀的濃度,同時(shí)測(cè)定全血樣本中血糖水平,并用DAS3.2.4軟件對(duì)試驗(yàn)數(shù)據(jù)進(jìn)行處理,計(jì)算藥動(dòng)學(xué)參數(shù)。結(jié)果多次口服沙格列汀片5 mg qd的主要藥動(dòng)學(xué)參數(shù)ρmax為(16.27±9.28)μg·L-1,tmax為(1.70±0.48)h,t1/2為(3.33±0.59)h,AUC0-12為(45.19±18.67)μg·h·L-1,AUC0-∞為(49.15±19.71)μg·h·L-1。沙格列汀的血藥濃度與血糖水平無(wú)特異相關(guān)性。結(jié)論沙格列汀應(yīng)用于精神分裂癥合并2型糖尿病患者藥動(dòng)學(xué)特征在正常范圍內(nèi),安全性較好,對(duì)于部分血糖波動(dòng)較大的患者控制血糖能力一般。
