塞來昔布對顱腦創(chuàng)傷后Caspase-9表達(dá)及運(yùn)動功能的影響

【摘要】:目的探討塞來昔布對大鼠顱腦創(chuàng)傷后環(huán)氧合酶(COX-2)和凋亡蛋白Caspase-9表達(dá)及運(yùn)動功能的影響。方法實(shí)驗(yàn)分為對照組、假手術(shù)組、創(chuàng)傷組、治療組,采用Marmarou方法建立大鼠閉合性顱腦創(chuàng)傷模型,實(shí)時熒光定量PCR(q PCR)檢測COX-2和Caspase-9基因表達(dá)量,免疫組織化學(xué)染色法檢測COX-2和Caspase-9蛋白的表達(dá),神經(jīng)功能損害評分(NSS)檢測大鼠的運(yùn)動功能。結(jié)果創(chuàng)傷組COX-2和Caspase-9基因和蛋白的表達(dá)明顯高于其他3組(P0.05),治療組較創(chuàng)傷組能有效降低COX-2和Caspase-9的表達(dá),但仍高于假手術(shù)組和正常組(P0.05);與創(chuàng)傷組相比較,治療組能有效改善大鼠的運(yùn)動功能障礙(P0.05),和對照組和假手術(shù)組比較差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論塞來昔布可通過對COX-2的特異性抑制作用,減少顱腦損傷后炎癥反應(yīng),并進(jìn)一步降低Caspase-9的表達(dá),從而減少神經(jīng)細(xì)胞的凋亡,并改善大鼠顱腦創(chuàng)傷后的運(yùn)動功能障礙。
[Abstract]:Objective to investigate the effect of celecoxib on the expression of cyclooxygenase (COX-2) and apoptosis protein Caspase-9 after traumatic brain injury in rats. Methods the experimental group was divided into control group, sham operation group, trauma group and treatment group. The model of closed craniocerebral trauma was established by Marmarou method, and real-time fluorescence quantitative PCR (Q PCR) was used to detect COX-2 and Caspase-9 base. The expression of COX-2 and Caspase-9 protein was detected by immunohistochemical staining, and the motor function of rats was detected by nerve function impairment score (NSS). Results the expression of COX-2 and Caspase-9 gene and protein in the trauma group was significantly higher than that of the other 3 groups (P0.05). The treatment group was more effective than the trauma group to reduce the expression of COX-2 and Caspase-9, but it was still higher than that in the trauma group. The sham operation group and the normal group (P0.05), compared with the trauma group, the treatment group could effectively improve the motor dysfunction (P0.05) in the rats (P0.05). Conclusion the celecoxib could reduce the inflammatory reaction after the brain injury and further reduce the Caspas through the specific suppression of COX-2. The expression of e-9 can reduce neuronal apoptosis and improve motor dysfunction after traumatic brain injury in rats.
