SHR大鼠腎臟及循環(huán)中CD13表達(dá)水平與原發(fā)性高血壓發(fā)病關(guān)系的實(shí)驗(yàn)研究

【摘要】:目的:初步探討CD13分子在原發(fā)性高血壓疾病中的表達(dá)水平及其在高血壓發(fā)生發(fā)展中的作用。 方法:4周齡SPF級(jí)雄性自發(fā)性高血壓大鼠(spontaneously hypertensive rat,SHR)及其野生對(duì)照組京都維斯塔爾大鼠(Wistar-Kyoto rat, WKY)各32只,按簡(jiǎn)單隨機(jī)法將SHR、WKY大鼠各分為4組,每組8只,分別在4周(O期)、6周(高血壓起始期)、10周(高血壓成型期)、23周(靶器官損害期)進(jìn)行實(shí)驗(yàn)觀察。采用無創(chuàng)尾動(dòng)脈血壓測(cè)量?jī)x動(dòng)態(tài)監(jiān)測(cè)SHR、WKY的血壓情況;應(yīng)用ELISA法動(dòng)態(tài)檢測(cè)上述大鼠的24h尿蛋白情況;Trizol法提取各組大鼠腎臟組織總RNA,并逆轉(zhuǎn)錄為cDNA,RT-PCR法動(dòng)態(tài)檢測(cè)大鼠腎臟中CD13mRNA的表達(dá)水平;取各組大鼠腎臟皮質(zhì)髓質(zhì)縱切面2mm厚度組織,石蠟法常規(guī)脫水包埋連續(xù)切片,應(yīng)用免疫組織化學(xué)技術(shù)動(dòng)態(tài)檢測(cè)CD13蛋白的表達(dá)水平。應(yīng)用ELISA法動(dòng)態(tài)檢測(cè)CD13、AngⅢ、AngⅣ在大鼠血漿中的表達(dá)水平并分析上述指標(biāo)與SHR血壓變化的相關(guān)性;采用HE及MASSON染色檢測(cè)各期大鼠腎臟病理改變情況。 結(jié)果:SHR大鼠體重在觀察期間內(nèi)均低于同期野生對(duì)照WKY大鼠,在6周、10周、23周分別為(160.2±5.3)g vs.(186.8±6.8)g、(230.6±13.5)g vs.(252.3±5.5) g,(310.7±9.1) g vs.(357.4±7.0)g,兩組差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。SHR大鼠血壓值亦高于同期野生對(duì)照WKY大鼠,在6周、10周、23周分別為(128.8±10.3)mmHg vs.(109.1±3.8)mmHg、(152.6±8.6)mmHg vs. mmHg、(203.9±13.2) mmHg vs.(112.9±6.9)mmHg,兩組差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。RT-PCR結(jié)果顯示,SHR腎臟中CD13mRNA表達(dá)水平高于同期野生對(duì)照WKY大鼠。在6周、10周、23周分別為0.36+0.05vs.0.25+0.03、0.61±0.08vs.0.28±0.03、0.62±0.09vs.0.17±0.02,兩組差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。免疫組化染色結(jié)果顯示,SHR腎臟中CD13蛋白表達(dá)水平亦高于同期野生對(duì)照WKY大鼠。在6周、10周、23周分別為0.42±0.12vs.0.25±0.15、0.57±0.13vs.0.38±0.09、0.73±0.12vs.0.37±0.19,兩組差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。血漿中CD13的表達(dá)低于同期野生對(duì)照WKY大鼠。在10周、23周分別為(107.4±22.3) ng/L vs.(147.1±25.5) ng/L、(87.04±20.7)ng/L vs.(150.1±31.2)ng/L,兩組差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。此外,SHR大鼠血漿中CD13的變化值與血壓呈負(fù)相關(guān)(r=-0.758,P0.05);SHR大鼠血漿中AngⅢ的變化值與血壓呈正相關(guān)(r=0.711,P0.05); SHR大鼠血漿中AngⅣ的變化值與血壓呈負(fù)相關(guān)(r=-0.732,P0.05);且SHR大鼠血漿中CD13表達(dá)水平動(dòng)態(tài)變化與AngⅢ表達(dá)水平的動(dòng)態(tài)變化亦具有負(fù)相關(guān)性(r=-0.746, P0.05); SHR大鼠血漿中CD13表達(dá)水平動(dòng)態(tài)變化與AngⅣ表達(dá)水平的動(dòng)態(tài)變化亦具有正相關(guān)性(r=0.698,P0.05)。HE和MASSON染色結(jié)果顯示,SHR大鼠腎臟微血管周圍、腎小管間質(zhì)膠原沉積多于野生對(duì)照WKY大鼠,在10周、23周差異明顯。 結(jié)論:CD13在腎臟中和循環(huán)系統(tǒng)中可能具有不同的生物學(xué)功能,參與調(diào)控機(jī)體的血壓及其病理改變。
[Abstract]:Objective: to investigate the expression of CD13 in essential hypertension and its role in the development of hypertension.Methods Thirty two Wistar Kyoto (WKYY) rats (WKYY) and 32 Wistar Kyoto rats (WKYY) were randomly divided into 4 groups (8 rats in each group).The experimental observation was carried out at 4 weeks and 6 weeks (the onset of hypertension was 10 weeks) and the target organ damage stage was 23 weeks (target organ damage phase).The blood pressure of SHR-WKY was dynamically monitored by non-invasive caudal artery blood pressure meter.The 24 hours urine protein of the above mentioned rats was dynamically detected by ELISA method and the total RNAs of kidney tissue were extracted by Trizol method, and the expression of CD13mRNA in the kidney was dynamically detected by reverse transcriptase reverse transcription-polymerase chain reaction (RT-PCR), and the thickness of 2mm in the medulla longitudinalis of renal cortex of the rats in each group was obtained.Paraffin method was used to detect the expression of CD13 protein by immunohistochemical technique.The expression of CD13 Ang 鈪,
