不同發(fā)育階段的BX-SPF雞消化道內(nèi)分泌細(xì)胞的研究

【摘要】:本文應(yīng)用Grimelius法對(duì)BX-SPF雞11個(gè)發(fā)育階段(1天、3天、1周、2周、3周、4周、10周、21周、26周、40周、69周)的消化道嗜銀細(xì)胞進(jìn)行了研究。結(jié)果顯示,以上各發(fā)育階段雞消化道除食管外,其它部位均有嗜銀細(xì)胞分布,且細(xì)胞形態(tài)各異:有圓形、橢圓形、水滴形、錐體形和梭形等,嗜銀細(xì)胞分布在胃部腺葉區(qū),腸上皮細(xì)胞之間、腺泡上皮之間及固有膜內(nèi);對(duì)雞胃腸道同一部位不同發(fā)育階段嗜銀細(xì)胞的分布密度進(jìn)行多重比較,發(fā)現(xiàn)4周齡組胃腸道除了回腸以外,剩余部位均高于其它年齡組,且差異顯著;同一發(fā)育階段胃腸道不同部位嗜銀細(xì)胞相比,各年齡組腺胃和直腸均高于胃腸道其它部位,且差異極顯著。本文還應(yīng)用免疫組織化學(xué)ABC法(Avidin-biotin complex method,ABC)對(duì)以上各年齡段BX-SPF雞消化道5-羥色胺(5-Hydroxytryptamine,5-HT)細(xì)胞和胃泌素(Gastrin,Gas)細(xì)胞進(jìn)行免疫組織化學(xué)鑒別和定位。結(jié)果顯示,5-HT細(xì)胞在各個(gè)發(fā)育階段雞消化道中除食管外,其它部位均有分布(腺胃、小腸、直腸和盲腸);Gas細(xì)胞只存在于各年齡中的小腸(十二指腸、空腸和回腸)部位。2種內(nèi)分泌細(xì)胞形態(tài)相似,均有圓形、橢圓形、水滴形、錐體形和少量的梭形等,但分布位置有差異,5-HT細(xì)胞在胃部主要分布于腺葉區(qū),在腸部主要分布于上皮細(xì)胞之間、腺泡上皮之間及固有膜內(nèi);而胃泌素細(xì)胞主要分布于腸上皮細(xì)胞之間、固有膜內(nèi),少數(shù)分布在腺泡上皮之間偶見。對(duì)雞消化道同一部位不同發(fā)育階段的5-HT細(xì)胞及Gas細(xì)胞分別進(jìn)行多重比較,發(fā)現(xiàn)26周齡BX-SPF雞胃腸道5-HT細(xì)胞與其它年齡段相比除了直腸外,均高于其它年齡組,且有顯著差異,Gas細(xì)胞分布密度呈現(xiàn)波浪形變化。5-HT細(xì)胞在同一發(fā)育階段胃腸道不同部位之間相比,腺胃部分布密度明顯低于胃腸其它部位,而直腸部密度分布又明顯高于胃腸其它部位,且差異顯著;Gas細(xì)胞在同一年齡組小腸不同部位之間相比,回腸部均高于十二指腸和空腸(除1天、3天和1周組外)。以上不同發(fā)育階段消化道內(nèi)分泌細(xì)胞形態(tài)、位置以及密度分布特征與雞出生后各個(gè)不同時(shí)期生長(zhǎng)發(fā)育規(guī)律以及消化道不同部位生理功能是相適應(yīng)的。
[Abstract]:In this paper, Grimelius method was used to study the silver eosinophils in the digestive tract of BX-SPF chickens in 11 developmental stages (1 day, 3 days, 1 week, 2 weeks, 4 weeks, 10 weeks, 21 weeks, 26 weeks, 40 weeks, 40 weeks, 69 weeks). The results showed that the digestive tract of the above developed chickens was in addition to the esophagus. In other parts, the distribution of argyrophilic cells was different: round, oval, water droplet, pyramidal and fusiform, and the silver cells were distributed in the glandular lobes of the stomach, between the intestinal epithelial cells, between the acinar epithelium and in the lamina propria. The distribution density of argyrophilic cells in the same part of the gastrointestinal tract in different stages of development was compared. The results showed that the distribution density of the silver eosinophils in the gastrointestinal tract of 4 weeks old was higher than that in the other age groups except the ileum, and the difference was significant. The adenogastric and rectum cells in different parts of the gastrointestinal tract in the same developmental stage were higher than those in the other parts of the gastrointestinal tract. The immunohistochemical identification and localization of 5-hydroxytryptamine 5-HT cells and gastrin gastrin gastrin gas cells in digestive tract of BX-SPF chickens of all ages were carried out by using the immunohistochemical ABC method Avidin-biotin complex method. The results showed that 5-HT cells were located in the digestive tract. In the digestive tract, except esophagus, There were only 2 endocrine cells in the small intestine (duodenum, jejunum and ileum) in all the other sites (glandular stomach, small intestine, rectum and cecum), all of which were round, oval, and water droplet shape, but only in the small intestine (duodenum, jejunum and ileum). The distribution of 5-HT cells was mainly in the glandular lobes in the stomach, between the epithelial cells in the intestine, between the epithelial cells in the acinar epithelium and in the lamina propria. However, gastrin cells were mainly distributed between intestinal epithelial cells, in the lamina propria and a few in acinar epithelium. The 5-HT cells and Gas cells in the same part of the digestive tract were compared in different stages of development. It was found that 5-HT cells in gastrointestinal tract of 26 week old BX-SPF chickens were higher than those of other age groups except rectum. There was significant difference in the distribution density of Gas cells. The distribution density of 5-HT cells in the glandular stomach was significantly lower than that in the other parts of the gastrointestinal tract in the same developmental stage. The density distribution of rectum was significantly higher than that of other parts of gastrointestinal tract, and the difference was significant between different parts of small intestine in the same age group. The ileum was higher than duodenum and jejunum (except 1 day 3 days and 1 week group). The distribution characteristics of position and density were suitable for the growth and development of chicken at different stages after birth and the physiological function of different parts of digestive tract.
