金線蓮多糖對(duì)環(huán)磷酰胺所致免疫抑制小鼠脾臟細(xì)胞因子及轉(zhuǎn)錄因子mRNA表達(dá)的影響

【摘要】:50只雄性昆明小鼠適應(yīng)性飼養(yǎng)1周后隨機(jī)分為5組,分別為空白對(duì)照組、模型組及金線蓮多糖(ARP)低、中、高劑量組,每組10只;第1~3天,除空白對(duì)照組注射生理鹽水外,其他各組腹腔注射環(huán)磷酰胺(CTX)80mg/kg,連續(xù)3d;第4~10天,空白組和模型組灌服蒸餾水,其他組分別灌服100,200,400mg/kg的ARP。末次給藥24h后,采集脾臟,qRT-PCR檢測(cè)試驗(yàn)小鼠脾臟細(xì)胞因子(IL-2、IFN-γ、IL-4、IL-6)及轉(zhuǎn)錄因子(T-bet、GATA-3)mRNA表達(dá)。結(jié)果顯示:與空白對(duì)照組相比,模型組IL-2、GATA-3 mRNA表達(dá)量降低,IL-4、IL-6、IFN-γ、T-bet mRNA表達(dá)量升高;與模型組相比,ARP高劑量組小鼠IL-4mRNA相對(duì)表達(dá)量降低(P0.05),中、高劑量組IL-6、IFN-γmRNA相對(duì)表達(dá)量降低(P0.05),ARP組T-bet的相對(duì)表達(dá)量降低(P0.05),高劑量組GATA-3的相對(duì)表達(dá)量升高(P0.05)。結(jié)果表明:ARP通過上調(diào)或下調(diào)小鼠脾臟細(xì)胞因子與轉(zhuǎn)錄因子的表達(dá),調(diào)節(jié)免疫抑制小鼠的免疫功能。
[Abstract]:After one week of adaptive feeding, 50 male Kunming mice were randomly divided into 5 groups: blank control group, model group and AARP group with low, medium and high doses, 10 rats in each group. The other groups were intraperitoneally injected with CTX 80 mg / kg of cyclophosphamide for 3 days. On the 4th day, the blank group and the model group were given distilled water, and the other groups were given 100 ~ 200g / kg ARP of 400 mg / kg respectively. 24 hours after the last administration, The expression of IL-2mil and T-beta-GATA-3T mRNA were detected by qRT-PCR. The results showed that compared with the control group, the expression of IL-2tGATA-3 mRNA in the model group was significantly lower than that in the control group, and the expression of IL-4 IL-6IFN- 緯 T-bet mRNA in the model group was higher than that in the control group. Compared with the model group, the relative expression of IL-4mRNA in the high dose group was lower than that in the model group. The relative expression of T-bet was decreased in high dose group (P 0.05), and increased in high dose group (P 0.05). The results showed that the expression of spleen cytokines and transcription factors were up-regulated or down-regulated. To regulate the immune function of immunosuppressive mice.
