不同氮能比飼糧對(duì)妊娠環(huán)江香豬臍帶血管發(fā)育相關(guān)基因表達(dá)的影響

【摘要】:本試驗(yàn)旨在研究不同氮能比飼糧對(duì)妊娠環(huán)江香豬臍帶血管發(fā)育相關(guān)基因表達(dá)的影響。選用首次妊娠的環(huán)江香豬48頭,根據(jù)體重隨機(jī)分為2組,每組8個(gè)重復(fù)(欄),每個(gè)重復(fù)3頭豬。配種后分別飼喂高氮能比飼糧[消化能(DE)為14.73 MJ/kg,粗蛋白質(zhì)(CP)含量為13.11%,氮能比為0.89]和低氮能比飼糧(DE為12.24 MJ/kg,CP含量為9.77%,氮能比為0.80)。分別于妊娠第45、75和110天每欄取1頭母豬,處死屠宰,收集每窩中最大體重、平均體重和最小體重胎豬對(duì)應(yīng)的臍帶,利用實(shí)時(shí)熒光定量PCR方法檢測(cè)血管發(fā)育相關(guān)基因——成纖維細(xì)胞生長(zhǎng)因子(FGF)、血管內(nèi)皮生長(zhǎng)因子(VEGF)、血管內(nèi)皮生長(zhǎng)因子受體2(VEGFR2)和內(nèi)皮型一氧化氮合酶(e NOS)的表達(dá)量。結(jié)果表明:與低氮能比飼糧組相比,高氮能比飼糧顯著上調(diào)了臍帶血管發(fā)育相關(guān)基因的表達(dá)量(P0.05),包括妊娠45 d最大體重胎豬的VEGF和VEGFR2表達(dá)量,平均體重胎豬的VEGF表達(dá)量;妊娠75 d最大體重胎豬的VEGF、VEGFR2和e NOS表達(dá)量,平均體重胎豬的VEGF和e NOS表達(dá)量,最小體重胎豬的VEGFR2表達(dá)量;妊娠110 d最大體重胎豬的FGF、VEGFR2和e NOS表達(dá)量,平均體重胎豬的FGF和VEGFR2表達(dá)量,最小體重胎豬的VEGF和e NOS表達(dá)量。不同妊娠期相同標(biāo)準(zhǔn)體重、同一妊娠期不同標(biāo)準(zhǔn)體重胎豬的臍帶血管發(fā)育相關(guān)基因的表達(dá)量均存在差異。由此可見(jiàn),高氮能比飼糧可能由于其蛋白質(zhì)和氨基酸水平較高而顯著上調(diào)了妊娠環(huán)江香豬臍帶血管發(fā)育相關(guān)基因的表達(dá)量,不同妊娠期或同一妊娠期不同標(biāo)準(zhǔn)體重胎豬的臍帶血管發(fā)育相關(guān)基因的表達(dá)量也存在差異。
[Abstract]:The purpose of this study was to study the effects of different nitrogen specific diets on the expression of genes related to umbilical cord blood tube development in pregnant Huanjiang Xiang pigs. 48 first pregnant Huanjiang Xiang pigs were randomly divided into 2 groups according to their body weight. There were 8 replicates in each group. After breeding, high nitrogen energy was 14.73 MJ / kg, crude protein CPR was 13.11, nitrogen energy ratio was 0.89 and low nitrogen energy ratio was 12.24 MJ / kg CP content, nitrogen energy ratio was 9.77%, nitrogen energy ratio was 12.24 MJ / kgCP, nitrogen energy ratio was 14.73 MJ / kg, crude protein / energy ratio was 13.11, and low nitrogen energy ratio was 12.24 MJ / kgCP, respectively. 1 sows per column on the 45th and 110th days of pregnancy, They were killed and slaughtered, and the umbilical cord corresponding to the maximum body weight, average body weight and minimum body weight of each litter were collected. The expression of vascular growth factor-fibroblast growth factor (FGFF), vascular endothelial growth factor (VEGF), vascular endothelial growth factor receptor 2 (VEGFR2) and endothelial nitric oxide synthase (Enos) were detected by real-time fluorescence quantitative PCR. The results showed that: compared with the low nitrogen energy group, Compared with diet, high nitrogen energy significantly upregulated the expression of cord blood vessel development related genes (P0.05), including the expression of VEGF and VEGFR2 in the maximum body weight fetal pigs at 45 days of gestation, and the average VEGF expression in the average body weight fetal pigs, and the VEGFFR2 and e NOS expression in the maximum body weight fetal pigs on the 75th day of gestation, and the expression of VEGFR2 and e NOS in the maximum body weight fetal pigs on the 75th day of gestation. The average expression of VEGF and e NOS, the expression of VEGFR2 in the least weight fetal pigs, the expression of VEGF VEGF FR2 and e NOS in the maximum body weight fetal pigs, and the expression of FGF and VEGFR2 in the average body weight fetal pigs. The expression levels of VEGF and e NOS in minimum weight fetal pigs were different. The expression levels of cord blood vessel development related genes in the same gestational period and the same standard weight fetal pigs were all different. The expression of genes related to the development of umbilical cord blood tube of pregnant Huanjiang Xiang pig was significantly up-regulated by high nitrogen energy compared with diet because of its high level of protein and amino acid. There were also differences in the expression of genes related to the development of umbilical blood vessel in different gestational period or the same gestational period of different standard weight fetal pigs.
