【摘要】： 在許多上皮來源的癌組織中,MUC1的表達(dá)上調(diào)、極性消失及糖基化程度降低使其成為多種腫瘤輔助診斷、指導(dǎo)治療及判斷預(yù)后的重要指標(biāo)。因而大量單克隆抗體被制備用于MUC1的檢測,我們決定自制抗人MUC1單克隆抗體(monoclonal antibody,McAb),建立ELISA體系檢測MUC1。首先采用大腸桿菌表達(dá)的8R-MUC1-6 His為免疫原制備McAb,用間接ELISA法和免疫組化檢測其效價和特異性。然以此得到的McAb無法滿足制備高靈敏度及高特異性ELISA檢測試劑盒的要求。為獲得高效價及高特異性的McAb,我們選用Bac-to-Bac桿狀病毒表達(dá)系統(tǒng)表達(dá)MUC1重組蛋白,此系統(tǒng)可以對其表達(dá)蛋白進(jìn)行簡單的糖基化,這一特點(diǎn)可使其更好的模擬腫瘤表面低糖基化的MUC1,可能成為制備McAb的理想抗原。
[Abstract]:In many epithelial cancer tissues, the up-regulated expression of MUC1, the disappearance of the polarity and the decrease of glycosylation make it an important indicator for multiple tumor assisted diagnosis, guiding treatment and judging the prognosis. Therefore, a large number of monoclonal antibodies have been prepared for the detection of MUC1, and we have determined the self-made anti human MUC1 monoclonal antibody (monoclonal antibody, McAb). The establishment of ELISA system to detect MUC1. first used 8R-MUC1-6 His expressed by Escherichia coli as immunogen to prepare McAb, using indirect ELISA and immunohistochemistry to detect its titer and specificity. However, the obtained McAb can not meet the requirements of preparing high sensitivity and high specificity ELISA detection kit. In order to obtain high efficient and highly specific McAb, I We use Bac-to-Bac baculovirus expression system to express the recombinant protein of MUC1. This system can make simple glycosylation of its expressed protein. This feature can make it better simulate the low glycosylated MUC1 of the tumor surface, and may be an ideal antigen for the preparation of McAb.
