巴西蘇木素調(diào)控自噬對(duì)高糖誘導(dǎo)血管內(nèi)皮細(xì)胞損傷的影響

【摘要】:目的探討巴西蘇木素對(duì)高糖誘導(dǎo)血管內(nèi)皮細(xì)胞損傷的保護(hù)作用及其可能機(jī)制,為中藥蘇木治療糖尿病大血管病變提供實(shí)驗(yàn)依據(jù)。方法培養(yǎng)人臍靜脈內(nèi)皮細(xì)胞,采用MTT法確定巴西蘇木素濃度,高糖誘導(dǎo)損傷后加入巴西蘇木素,透射電鏡觀察自噬體;小管形成實(shí)驗(yàn)觀察血管生成;蛋白免疫印跡檢測(cè)自噬相關(guān)基因BECLIN1、LC3-Ⅱ、p62蛋白的表達(dá)。結(jié)果選定用于實(shí)驗(yàn)的巴西蘇木素濃度為15 mg/L;透射電鏡顯示高糖組細(xì)胞中自噬體數(shù)量較對(duì)照組少,而巴西蘇木素組細(xì)胞中自噬體數(shù)量較對(duì)照組和高糖組多;BECLIN1、LC3-Ⅱ的蛋白表達(dá)在高糖組較對(duì)照組低,在巴西蘇木素組較高糖組高(P0.05);p62的蛋白表達(dá)在高糖組較對(duì)照組高,在巴西蘇木素組較對(duì)照組和高糖組低(P0.05)。結(jié)論巴西蘇木素可能通過調(diào)控自噬而對(duì)高糖誘導(dǎo)血管內(nèi)皮細(xì)胞損傷發(fā)揮保護(hù)作用。
[Abstract]:Objective to investigate the protective effect of Sappan Brazil high glucose induced vascular endothelial cell injury and its possible mechanism, to provide experimental basis for Chinese medicine sappan treatment of diabetic macrovascular disease. Methods human umbilical vein endothelial cells, determine the Brazil hematoxylin concentration using MTT method, high glucose induced injury after joining the Brazil hematoxylin, transmission electron microscopy observation of autophagosomes; experiment to observe the angiogenesis formation of tubules; Western blot detection of autophagy related gene BECLIN1, LC3- II, p62 protein expression was selected to experiment in Brazil. Hematoxylin concentration was 15 mg/L; transmission electron microscopy showed that the number of cells in the body of autophagy in high glucose group than in the control group, while the number of Brazil hematoxylin cells autophagy body than the control group and high glucose group; BECLIN1, the expression of LC3- II protein in high glucose group than the control group in Brazil, hematoxylin group compared with high glucose group (P0.05); high p62 protein expression in The high glucose group was higher than the control group, and lower in Brazil hematoxylin group than the control group and the high sugar group (P0.05). Conclusion Brazil hematoxylin may play a protective role in the high glucose induced vascular endothelial cell injury by regulating autophagy.
