【摘要】:為探明鴨腸炎病毒(Duck enteritis virus,DEV)感染鴨脾臟的轉(zhuǎn)錄組情況,本研究以DEV GZ株經(jīng)腿部肌肉接種50d鴨,于接種后66h采集鴨脾組織樣本,提取總RNA,采用Illumina HiSeq 2000TM對試驗(yàn)組和對照組樣品進(jìn)行測序,篩選DEV感染鴨脾臟組織的差異表達(dá)基因,對其進(jìn)行生物信息學(xué)GO功能分類和KEGG信號通路分析。結(jié)果顯示,差異表達(dá)基因共511個,其中表達(dá)上調(diào)312個,表達(dá)下調(diào)199個。GO功能分類結(jié)果顯示,差異表達(dá)基因主要涉及氧運(yùn)輸、整合素活化、補(bǔ)體激活等生物學(xué)過程,胞外區(qū)、血紅蛋白復(fù)合物、細(xì)胞外間隙等細(xì)胞組分,氧轉(zhuǎn)運(yùn)活動、氧結(jié)合、核糖體的結(jié)構(gòu)成分等分子功能。KEGG分析表明這些差異表達(dá)基因參與ECM受體相互作用、核糖體、CAMs、JAK-STAT信號通路、細(xì)胞因子和細(xì)胞因子受體相互作用、PPAR信號通路、神經(jīng)活性配體/受體相互作用信號通路。本研究為深入探究DEV與鴨脾臟組織互作、宿主抗病機(jī)制及相關(guān)功能基因的篩選奠定了基礎(chǔ)。
[Abstract]:In order to find out the transcriptional status of duck spleen infected with duck enteritis virus (Duck enteritis virus), DEV GZ strain was inoculated with leg muscle for 50 d, and the samples of duck spleen tissue were collected at 66 h after inoculation. Total RNAs were extracted, Illumina HiSeq 2000TM was used to sequence the samples of the experimental group and the control group, and the differentially expressed genes in the spleen tissue of the infected duck were screened. The differential expression genes were classified by bioinformatics go and analyzed by KEGG signal pathway. There were 511 differentially expressed genes, including 312 up-regulated genes, 199 down-regulated .go functional classification results showed that differentially expressed genes mainly involved in oxygen transport, integrin activation, complement activation and other biological processes, extracellular domain, etc. Molecular functions of hemoglobin complex, extracellular intercellular components, oxygen transport, oxygen binding, ribosomal structural components, etc. KEGG analysis showed that these differentially expressed genes were involved in the interaction of ECM receptor, and ribosomal CAMsJAK-STAT signaling pathway. The interaction between cytokine and cytokine receptor is the signal pathway of PPAR and the signal pathway of neuroactive ligand / receptor interaction. The purpose of this study was to explore the interaction between DEV and duck spleen. The screening of host disease resistance mechanism and related functional genes laid the foundation.
