烏雞源芽孢桿菌與中草藥配伍對(duì)烏雞生長(zhǎng)性能與免疫性能的影響

【摘要】:益生芽孢桿菌作為抗生素替代品的研究很多,但關(guān)于其與復(fù)方中草藥配伍的研究卻很少,尤其是應(yīng)用于烏雞的研究幾乎沒(méi)有。本試驗(yàn)以珍貴藥禽——烏雞為試驗(yàn)對(duì)象,研究烏雞源芽孢桿菌與中草藥配伍對(duì)烏雞生長(zhǎng)與免疫性能的影響,旨在為新型抗生素替代品——烏雞源芽孢桿菌與中草藥配伍應(yīng)用于烏雞養(yǎng)殖提供理論依據(jù)。本試驗(yàn)從健康烏雞胃腸道篩選到2株芽孢桿菌WJ1、WJR2,分別對(duì)其進(jìn)行群體形態(tài)與個(gè)體形態(tài)觀察、生理生化試驗(yàn)等,并以16S r DNA序列分析構(gòu)建系統(tǒng)發(fā)育樹(shù),確定其系統(tǒng)發(fā)育地位:分離株WJ1判定為蠟樣芽胞桿菌,分離株WJR2判定為枯草芽孢桿菌。對(duì)分離到的WJ1、WJR2進(jìn)行產(chǎn)酶特性(淀粉酶、蛋白酶)、耐受性(人工胃液、人工腸液、豬膽鹽),以及培養(yǎng)特性研究,確定其均為高產(chǎn)酶與高耐受菌株;其培養(yǎng)學(xué)特性為:最適p H均為6,最適生長(zhǎng)溫度均為37℃左右,其中菌株WJ1溫度生長(zhǎng)范圍稍廣;生長(zhǎng)曲線(xiàn)特征為:兩菌均在2h~12h達(dá)到對(duì)數(shù)期,WJ1的生長(zhǎng)穩(wěn)定期在12h~34h;WJR2的生長(zhǎng)穩(wěn)定期在12h~24h。將WJ1、WJR2分別與0.5g、1g、1.5g三個(gè)不同劑量的復(fù)方中草藥(由黨參、黃芪、茯苓等按照一定比例組成)配伍進(jìn)行動(dòng)物益生性試驗(yàn),結(jié)果顯示:不同配伍均能夠不同程度提高烏雞日增重,降低料重比,促進(jìn)脾臟、法氏囊免疫器官的生長(zhǎng)發(fā)育、保持雞新城疫血凝抑制抗體在較高水平,還能有效的提高烏雞體內(nèi)免疫球G蛋白(Ig G)的含量。其中試驗(yàn)組Ⅳ(WJ1與1g復(fù)方中草藥配伍)在日增重、提高脾臟指數(shù)、維持ND效價(jià)在較高水平效果最為明顯。與對(duì)照組相比,日增重最多可達(dá)23.6%(P0.05);脾臟免疫指數(shù)42d時(shí)最高達(dá)29.1%(P0.05),ND效價(jià)最高達(dá)60%(P0.05)。試驗(yàn)組Ⅵ(WJR2與0.5g復(fù)方中草藥配伍)料重比降低最為明顯,36-42d最多可降低19.1%(P0.05)。試驗(yàn)組Ⅷ(WJR2與1.5g復(fù)方中草藥配伍)提高法氏囊免疫指數(shù)、IgG含量效果最為明顯。與對(duì)照組相比,法氏囊免疫指數(shù)35d時(shí)最高達(dá)50.5%(P0.05);Ig G含量28d最高為50.5%(P0.05)。
[Abstract]:Bacillus probioticus as a substitute for antibiotics has been studied a lot, but few studies have been done on its compatibility with compound Chinese herbal medicine, especially on silky chicken. To study the effect of compatibility of Bacillus spp and Chinese herbal medicine on growth and immune performance of silky chicken. The aim of this study was to provide a theoretical basis for the application of a new antibiotic substitute, Bacillus from silky chicken, in combination with Chinese herbal medicine in silky chicken culture. In this study, two strains of Bacillus sp. WJ1 and WJR2 were screened from gastrointestinal tract of healthy silky chicken, and their population morphology was studied. And individual morphological observation, The phylogenetic tree was constructed by 16s r DNA sequence analysis. The phylogenetic status of the phylogenetic tree was determined. The isolated strain WJ1 was identified as Bacillus cereus. The isolated strain WJR2 was identified as Bacillus subtilis. The enzyme producing characteristics (amylase, protease, tolerance (artificial gastric juice, artificial intestinal fluid, porcine bile salt), and culture characteristics of the isolated WJ1 / WJR2 were determined to be high enzyme and high tolerance strains.
