【摘要】：目的:研究扶正康復(fù)合劑對移植性肺癌的抑制作用,并探討其作用機制。方法:將48只NOD/SCID裸鼠隨機分為8組:模型對照組,順鉑組,扶正康復(fù)合劑低、中、高劑量組,順鉑+扶正康復(fù)合劑低、中、高劑量組,共8組,每組6只,皮下接種H1975細(xì)胞建立移植性裸鼠肺癌模型,接種15 d后開始給藥,連續(xù)給藥7 d,稱取體重及瘤重,計算抑瘤率,ELISA法檢測裸鼠血清BUN、Cr、TNF-α、IFN-γ、IL-2、IL-6的表達(dá)水平,免疫組化法檢測瘤體組織VEGF、Bcl-2、Bax、Survivin的表達(dá)情況,q-PCR法檢測瘤體組織VEGF、Bcl-2、Bax、Survivin mRNA的表達(dá)水平。結(jié)果:扶正康復(fù)合劑高劑量組、順鉑組、順鉑+扶正康復(fù)合劑低、中、高劑量組與模型對照組相比:荷瘤裸鼠瘤質(zhì)量顯著減輕(P0.05),血清TNF-α水平顯著升高(P0.05),IL-2水平顯著升高(P0.05),血清IL-6水平顯著降低(P0.05),Bcl-2表達(dá)量顯著減少,Bax表達(dá)量增加。其中順鉑+扶正康復(fù)合劑高劑量給藥組裸鼠抑瘤率最高,兩者具有協(xié)同作用。與順鉑組相比:順鉑+扶正康復(fù)合劑高劑量組裸鼠血清Cr、BUN水平顯著降低(P0.01),順鉑+扶正康復(fù)合劑中、高劑量組裸鼠血清TNF-α、INF-γ水平顯著升高(P0.01);順鉑+扶正康復(fù)合劑高劑量組Bcl-2、Survivin、VEGF表達(dá)量降低,Bax表達(dá)量增加。結(jié)論:扶正康復(fù)合劑與順鉑聯(lián)用能夠降低順鉑的腎損傷作用,調(diào)節(jié)機體的Th1/Th2細(xì)胞免疫平衡,通過抑制Bcl-2、Survivin、VEGF并促進(jìn)Bax的表達(dá),進(jìn)而促進(jìn)肺癌細(xì)胞的凋亡。
[Abstract]:Objective: to study the inhibitory effect of Fuzhenghuangji on transplanted lung cancer and its mechanism. Methods: 48 NOD/SCID nude mice were randomly divided into 8 groups: model control group and cisplatin group. The model of transplanted nude mice lung cancer was established by inoculating H1975 cells subcutaneously into 8 groups of low, middle and high dosage groups of low, medium and high dose of Fuzheng Kangfeng mixture and low, middle and high dose groups of Cisplatin Fuzheng Kangfeng mixture. After 15 days of inoculation, the drug was administered continuously for 7 days. The weight and tumor weight were weighed. The inhibition rate of tumor was calculated and Elisa was used to detect IL-2 in nude mice serum BUNCU TNF- 偽 and IFN- 緯. The expression of IL-6 was detected by immunohistochemical method. The expression of survivin was detected by immunohistochemical method. VEGF was detected by q-PCR. Results: high dose group, cisplatin group, cisplatin group, cisplatin Fuzheng convalescent mixture were low and medium. Compared with the model control group, the tumor quality of the high dose group decreased significantly (P 0.05), and the serum TNF- 偽 level increased significantly (P 0.05). The level of IL-2 was significantly higher than that of P0.05, and the level of serum IL-6 was significantly lower than that of P0.05, and the expression of Bcl-2 was significantly decreased. The expression of Bax was increased. The tumor inhibition rate of nude mice was the highest in the high dose group. Compared with the cisplatin group, the serum Cr of the nude mice in the high dose group was higher than that in the cisplatin Fuzheng Kang mixture group. The level of BUN decreased significantly (P 0.01). In Cisplatin Fuzheng Rehabilitation mixture, the serum TNF- 偽 / INF- 緯 level in high dose group increased significantly (P 0.01). The expression of VEGF was decreased in the high dose group of cisplatin Fuzheng convalescent mixture. Conclusion: the combination of Fuzhenghuangji and cisplatin can reduce the renal injury of cisplatin and regulate the immune balance of Th1/Th2 cells by inhibiting Bcl-2. Survivin VEGF also promotes the expression of Bax and promotes the apoptosis of lung cancer cells.
