有氧運(yùn)動(dòng)對(duì)大鼠大腦皮質(zhì)和海馬組織Wnt/β-Catenin信號(hào)通路的影響

目的:探討有氧運(yùn)動(dòng)對(duì)大鼠大腦皮質(zhì)和海馬組織Wnt/β-Catenin信號(hào)通路的影響,為揭示有氧運(yùn)動(dòng)改善神經(jīng)系統(tǒng)功能提供一定的理論基礎(chǔ)。方法:雄性6周齡SD大鼠(n=40)隨機(jī)分為安靜對(duì)照組(CG組,n=20)和運(yùn)動(dòng)組(EG組,n=20)。EG組大鼠進(jìn)行為期8周的跑臺(tái)訓(xùn)練。最后一次訓(xùn)練結(jié)束后48小時(shí),分離所有大鼠大腦皮質(zhì)和海馬組織。通過(guò)熒光定量PCR和Western blot檢測(cè)各組大鼠大腦皮質(zhì)和海馬組織中Wnt、β-Catenin、LRP5、LRP6、Axin1、CK1以及GSK3βmRNA和蛋白含量及其磷酸化水平。結(jié)果:經(jīng)過(guò)8周跑臺(tái)訓(xùn)練后,EG組大鼠大腦皮質(zhì)和海馬組織Wnt1和Wnt3兩種亞型mRNA和蛋白含量均顯著高于CG組;β-Catenin mRNA水平雖未發(fā)生顯著變化,但其蛋白含量卻顯著高于CG組,同時(shí)其Ser33/Ser37和Thr41/Ser45位點(diǎn)的磷酸化水平顯著低于CG組。EG組LRP5和LRP6 mRNA和蛋白含量均顯著高于CG組;而Axin1和CK1 mRNA和蛋白含量均顯著低于CG組。EG組GSK3βmRNA和蛋白含量并未發(fā)生顯著改變,但其Ser9位點(diǎn)的磷酸化水...
