釀酒葡萄皮渣對(duì)單欄飼養(yǎng)方式下公綿羊繁殖性能及睪丸抗氧化性的影響

【摘要】:旨在研究日糧中添加一定量的釀酒葡萄皮渣對(duì)綿羊繁殖性能及睪丸抗氧化性能的影響。試驗(yàn)選取30只5月齡,體重(25±1)kg的杜泊×小尾寒羊雜交公羊,采用完全隨機(jī)設(shè)計(jì)平均分為5組。1組自由活動(dòng),其余4組單欄飼養(yǎng),建立應(yīng)激模型。自由活動(dòng)綿羊飼喂未添加葡萄皮渣日糧,單欄飼養(yǎng)綿羊分別飼喂不同葡萄皮渣添加水平(0%、5%、10%、20%)日糧。試驗(yàn)期80d。試驗(yàn)結(jié)束后,屠宰取樣,測(cè)定睪丸系數(shù)并作附睪精子分析,測(cè)定睪丸組織抗氧化水平,對(duì)睪丸組織中Cu-ZnSOD、CAT、GPx4及Nrf2mRNA和蛋白相對(duì)表達(dá)量進(jìn)行研究。結(jié)果表明,與自由活動(dòng)組相比,單欄組綿羊睪丸組織中MDA及ROS水平升高(P0.01),睪丸重量顯著降低(P0.05),睪丸系數(shù)也表現(xiàn)出下降趨勢(shì)(P=0.06),附睪精子密度及頂體完整率顯著降低(P0.05),精子活動(dòng)率極顯著降低(P0.01),精子畸形率極顯著升高(P0.01),表明單欄飼養(yǎng)方式對(duì)綿羊造成氧化應(yīng)激,并在一定程度上影響綿羊繁殖性能。適量添加釀酒葡萄皮渣后,綿羊附睪精子密度和精子活動(dòng)率均顯著升高(P0.05),精子畸形率顯著降低(P0.05),睪丸組織MDA含量顯著下降(P0.01),SOD、CAT及GSH-Px活性均顯著提高(P0.01,P0.05,P0.01);睪丸組織中Cu-ZnSOD mRNA相對(duì)表達(dá)量顯著提高(P0.05),Cu-ZnSOD、CAT、GPx4蛋白相對(duì)表達(dá)量顯著提高(P0.05)。綜上表明,釀酒葡萄皮渣可以提高單欄飼養(yǎng)方式下綿羊睪丸組織抗氧化性,進(jìn)而改善綿羊繁殖性能。
[Abstract]:The purpose of this study was to study the effect of adding a certain amount of wine grape skin residue on the reproductive performance and testicular antioxidant performance of sheep. Thirty 5-month-old Dupo 脳 small-tailed Han sheep crossbred rams weighing (25 鹵1) kg were randomly divided into 5 groups. Group 1 was free to move, and the other four groups were fed in a single column to establish stress model. The free moving sheep were fed with diet without grape skin residue, and the sheep fed with different levels of grape skin residue (0%, 5%, 10%, 20%) were fed with different levels of grape skin residue (0%, 5%, 10%, 20%). The experimental period is 80 days. At the end of the experiment, the testicular coefficient was measured and the epididymal sperm was analyzed. The antioxidant level of testicular tissue was measured, and the relative expression of Cu-ZnSOD,CAT,GPx4, Nrf2mRNA and protein in testicular tissue was studied.
