飼糧高鐵對肉仔雞十二指腸黏膜鐵轉(zhuǎn)運(yùn)載體基因表達(dá)及組織微量元素含量的影響

【摘要】:本試驗旨在研究飼糧鐵含量對肉仔雞組織重要微量元素鐵、錳、銅、鋅含量及十二指腸黏膜主要鐵轉(zhuǎn)運(yùn)載體基因表達(dá)的影響,探討鐵對肉仔雞微量元素吸收和代謝的影響及其機(jī)制。將336只1日齡商品代羅斯308肉公雛按照體重隨機(jī)分成4個組,每組6個重復(fù),每個重復(fù)14只雞。對照組飼喂不額外添加鐵的基礎(chǔ)飼糧(實測鐵含量為78 mg/kg),鐵添加組分別飼喂以七水硫酸亞鐵(FeSO_4·7H_2O)形式添加100、250或500 mg/kg鐵的試驗飼糧(實測鐵含量分別為166、308和579 mg/kg)。試驗期21 d。各組試雞分別于7、14和21日齡屠宰分析肝臟、心臟、胰腺、十二指腸黏膜和脛骨灰中鐵、錳、銅、鋅含量及十二指腸黏膜中二價金屬轉(zhuǎn)運(yùn)蛋白(DMT1)和膜鐵轉(zhuǎn)運(yùn)蛋白(FPN1)mRNA表達(dá)水平。結(jié)果表明:1)500 mg/kg鐵添加組1~7日齡和8~14日齡的平均日增重顯著低于其他3組(P0.10),250和500 mg/kg鐵添加組1~7日齡的平均日采食量顯著低于其他2組(P0.10)。2)飼糧鐵含量對肉仔雞7、14、21日齡的血漿總鐵結(jié)合力以及全血血紅蛋白濃度(7日齡除外)和紅細(xì)胞壓積均無顯著影響(P0.10),但顯著影響7、14和21日齡血漿鐵含量和鐵飽和度(P0.10),二者均隨飼糧鐵含量增加而升高。3)7和14日齡心臟及7、14和21日齡肝臟、十二指腸黏膜、胰腺和脛骨灰鐵含量均隨飼糧鐵含量的增加而升高,7、14和21日齡十二指腸黏膜、胰腺和脛骨灰錳含量均隨飼糧鐵含量的增加而降低;飼糧添加鐵顯著降低7日齡胰腺鋅含量(P0.10),但對其他日齡胰腺和各日齡其他所測組織鋅含量以及各日齡所測各組織銅含量均無顯著影響(P0.10)。4)飼糧鐵含量顯著影響7、14和21日齡十二指腸黏膜DMT1和FPN1 mRNA表達(dá)水平(P0.10),各日齡DMT1和FPN1mRNA表達(dá)水平均隨飼糧鐵含量的增加而降低。以上結(jié)果提示,高鐵飼糧可能通過調(diào)控十二指腸黏膜DMT1和FPN1基因的表達(dá)降低錳和鋅在腸道的吸收,進(jìn)而減少錳和鋅在組織中的沉積。
[Abstract]:The purpose of this study was to investigate the effects of dietary iron content on the expression of iron, manganese, copper, zinc and major iron transporter genes in duodenal mucosa of broilers. To investigate the effect of iron on the absorption and metabolism of trace elements in broilers, 336 1-day-old broilers were randomly divided into 4 groups according to their body weight, 6 replicates in each group. The control group was fed a basal diet (the measured iron content was 78 mg / kg 路kg ~ (-1)) and the iron supplementation group was fed with FeSO _ 4 路7H _ 2O in the form of Fe _ 2O _ 4 路7H _ 2O (measured iron content). They were 166,308 mg / kg and 579 mg / kg 路kg ~ (-1), respectively. The liver was slaughtered and analyzed at the age of 7 ~ 14 and 21 days respectively during the trial period of 21 days. Iron, manganese, copper in heart, pancreas, duodenal mucosa and tibial ash, Zinc content, divalent metal transport protein (DMT1) and membrane iron transport protein (FPN 1) mRNA expression in duodenal mucosa. The results showed that the average daily gain (ADG) was significantly lower than that in the other three groups at the age of 7 days and 8 ~ 14 days after addition of 1: 1 ~ 500 mg/kg iron group. The results showed that the average daily gain of the three groups was significantly lower than that of the other three groups (P _ (0.10) O _ (250) and P _ (500) mg/kg). The average daily feed intake of the addition group was significantly lower than that of the other two groups (P 0.10 路2) the total iron binding capacity of plasma and the whole blood hemoglobin concentration and hematocrit were not significantly affected by the dietary iron content in the 7 ~ (14) ~ (21) day old broilers except for the 7 ~ (th) day) and the hematocrit (P _ (0.10)). However, the plasma iron content and iron saturation were significantly affected at 7 ~ 14 and 21 days of age. Both of them increased with the increase of dietary iron content. The heart of 7 and 14 days of age and liver of 7 ~ 14 and 21 days of age were increased with the increase of dietary iron content. The contents of gray iron in duodenal mucosa, pancreas and tibia increased with the increase of dietary iron content. The contents of gray manganese in pancreas and tibia decreased with the increase of dietary iron content. Dietary iron supplementation significantly decreased the zinc content of the pancreas at 7 days of age (P 0.10), but had no significant effect on the zinc content of the other day pancreas and other tissues and copper content of the tissues measured at each day of age. The expression level of DMT1 and FPN1mRNA in duodenal mucosa of 21 days old was P0.10, and the expression level of DMT1 and FPN1mRNA decreased with the increase of dietary iron content. High iron diet may reduce the absorption of manganese and zinc in intestine by regulating the expression of DMT1 and FPN1 genes in duodenal mucosa, and then decrease the deposition of manganese and zinc in tissues.
