白念珠菌生物被膜形成相關(guān)基因的功能研究

【摘要】:生物被膜形成是近年來(lái)發(fā)現(xiàn)的白念珠菌耐藥新機(jī)制。本課題首先利用激光共聚焦顯微鏡確證Farnesol對(duì)白念珠菌生物被膜形成的抑制作用,隨后采用cDNA芯片雜交法獲得經(jīng)Farnesol處理與未經(jīng)處理生物被膜的差異表達(dá)基因,并以水—烴兩相分離實(shí)驗(yàn)證明細(xì)胞表面疏水性下降是生物被膜形成受抑制的機(jī)制之一。將上述差異表達(dá)基因與其它白念珠菌生物被膜形成及耐藥相關(guān)芯片雜交實(shí)驗(yàn)得到的差異表達(dá)基因作比較,并經(jīng)克隆的全長(zhǎng)測(cè)序及生物信息學(xué)分析,得到一全長(zhǎng)基因THI13。采用“Ura-Blaster”技術(shù)敲除白念珠菌中的THI13基因,并進(jìn)行表型分析。結(jié)果表明,THI13基因缺失可導(dǎo)致白念珠菌條件性的菌絲形成抑制以及對(duì)小鼠毒力的顯著降低。
[Abstract]:Biofilm formation is a new mechanism of drug resistance found in Candida albicans in recent years. In this study, the inhibitory effect of Farnesol on biofilm formation of Candida albicans was confirmed by laser confocal microscopy, and then the differentially expressed genes were obtained by cDNA chip hybridization between treated and untreated biofilms. The hydrophobicity of cell surface was proved to be one of the mechanisms of inhibition of biofilm formation. The differentially expressed genes were compared with the differentially expressed genes obtained from other Candida albicans biofilm formation and drug-resistance-related microarray hybridization, and a full-length gene THI13was obtained by sequencing and bioinformatics analysis. THI13 gene from Candida albicans was knocked out by Ura-Blaster technique and phenotypic analysis was carried out. The results showed that the deletion of THI13 gene could result in the inhibition of conditioned mycelium formation and the decrease of toxicity to mice in Candida albicans.
