電離輻射對小鼠巨噬細(xì)胞Toll信號傳導(dǎo)通路的影響

【摘要】:免疫細(xì)胞間或細(xì)胞與介質(zhì)間相互識別的物質(zhì)基礎(chǔ)是免疫細(xì)胞膜分子,包括細(xì)胞表面的多種抗原、受體和其它分子。研究電離輻射對免疫細(xì)胞膜分子的作用,有助于深入了解輻射免疫效應(yīng)的本質(zhì),為臨床應(yīng)用放射治療脊柱惡性腫瘤提供相應(yīng)的理論依據(jù)。 本研究在以往研究的輻射免疫效應(yīng)機(jī)制基礎(chǔ)上,進(jìn)一步探討電離輻射全身照射后,小鼠腹腔巨噬細(xì)胞TLRs 介導(dǎo)信號傳導(dǎo)通路的機(jī)制。不同劑量全身照射小鼠后,采用流式細(xì)胞術(shù)檢測小鼠腹腔巨噬細(xì)胞Toll 信號通路上跨膜受體CD14、TLR4 以及細(xì)胞內(nèi)轉(zhuǎn)接蛋白MyD88 表達(dá)的變化,采用ELISA 法檢測效應(yīng)產(chǎn)物IL-18表達(dá)的變化。結(jié)果顯示高低劑量X 射線全身照射小鼠后,均能增強(qiáng)該信號通路上的跨膜受體、轉(zhuǎn)接蛋白以及效應(yīng)產(chǎn)物的表達(dá)。說明電離輻射效應(yīng)影響小鼠機(jī)體免疫應(yīng)答,并且導(dǎo)致腹腔巨噬細(xì)胞的免疫因子IL-18 分泌增高,這是通過腹腔巨噬細(xì)胞固有免疫Toll信號傳導(dǎo)通路,即細(xì)胞膜上的受體CD14、TLR4/TLRs→胞內(nèi)轉(zhuǎn)接蛋白MyD88→細(xì)胞核轉(zhuǎn)錄因子NF-κB→效應(yīng)因子IL-18 來完成的。
[Abstract]:The material basis for mutual recognition between immune cells or between cells and mediators is the immune cell membrane molecules, including multiple antigens, receptors and other molecules on the surface of the cell. It is helpful to understand the nature of radio-immune effect and to provide theoretical basis for clinical radiotherapy of spinal malignant tumors. In this study, the mechanism of TLRs mediated signal transduction by peritoneal macrophages in mice was further studied on the basis of the previous studies on the mechanism of radiation immunity. Flow cytometry was used to detect the expression of transmembrane receptor CD14TLR4 and MyD88 on the Toll signaling pathway of mouse peritoneal macrophages. ELISA method was used to detect the expression of IL-18. The results showed that the transmembrane receptors in the signaling pathway were enhanced in mice exposed to high and low doses of X-rays. The expression of transfer protein and its effector products. The results show that ionizing radiation effects the immune response of mice and leads to the increase of IL-18 secretion of peritoneal macrophages, which is through the intrinsic immune Toll signal transduction pathway of peritoneal macrophages.
