雌二醇和孕酮對小鼠心肌細(xì)胞超微結(jié)構(gòu)的影響

【摘要】:目的 通過觀察苯甲酸雌二醇、乙酸羥孕酮以及二者聯(lián)用替代療法短期和長期作用對卵巢切除小鼠心肌細(xì)胞的數(shù)量、超微結(jié)構(gòu)形態(tài)學(xué)改變,從而探討苯甲酸雌二醇、乙酸羥孕酮對機體心血管系統(tǒng)的作用機制。 方法 采用健康雌性昆明小鼠60只,隨機分為五組:一組為保留卵巢空白對照組(INT),未加任何處理;其余四組去勢后,分別注射生理劑量的苯甲酸雌二醇(OVX+EB組)、乙酸羥孕酮(OVX+HPC組)、苯甲酸雌二醇、乙酸羥孕酮聯(lián)用(OVX+EB.HPC組)以及麻油(麻油對照組,OVX組)。每組12只,藥物注射從小鼠40天開始,每5天注射藥物一次,30次為一個療程,共150天。分別與半個療程和療程結(jié)束時各組取材制備電鏡標(biāo)本,代表短期治療組和長期治療組,電鏡下觀察各組心肌細(xì)胞超微結(jié)構(gòu)的形態(tài)學(xué)改變。 結(jié)果 不同治療組間比較:形態(tài)學(xué)觀察:空白對照組(INT)電鏡下心肌細(xì)胞超微結(jié)構(gòu)未見改變,符合正常心肌的組織形態(tài)。雌二醇組(OVX+EB)、孕酮組(OVX+HPC)、雌二醇加孕酮組(0VX+EB.HPC)和去卵巢組(OVX)心肌細(xì)胞超微結(jié)構(gòu)有不同程度的改變。去卵巢組與對照組比較改變最為明顯,以凝集性改變?yōu)橹。雌二醇組、孕酮組、雌二醇加孕酮組超微結(jié)構(gòu)改變程度介于對照組與去卵巢組之間。取閏盤形態(tài)變化作定性指標(biāo)進(jìn)行統(tǒng)計學(xué)處理,短期治療組與長期治療組統(tǒng)計結(jié)果顯示:OVX組與其它四組比較差別有統(tǒng)計學(xué)意義(P0.01);OVX+EB組、OVX+HPC組和OVX+EB.HPC組與INT組比較差別沒有統(tǒng)計學(xué)意義(P0.05);OVX+EB組、OVX+HPC組和OVX+EB.HPC組與OVX組比較差別有統(tǒng)計學(xué)意義(P0.01);OVX+EB組、OVX+HPC組和
[Abstract]:Objective to investigate the morphologic changes of cardiomyocytes in ovariectomized mice by observing the effects of estradiol benzoate, hydroxyprogesterone acetate and the combination of them on the number and ultrastructure of cardiomyocytes in ovariectomized mice. Mechanism of Action of Hydroxyprogesterone Acetate on Cardiovascular system. Methods Sixty healthy female Kunming mice were randomly divided into five groups: one group was treated without any treatment, the other four groups were ovariectomized. 12 rats in each group were injected with estradiol benzoate OVX EB group, OVX HPC group with hydroxyprogesterone acetate, estradiol benzoate and hydroxyprogesterone acetate combined with OVX EB.HPC group, and sesame oil with 12 rats in each group. Every 5 days, 30 times of drug injection were used as a course of treatment for 150 days. Electron microscopic specimens were taken from each group at the end of half course and treatment course respectively, representing the short-term treatment group and the long-term treatment group. The ultrastructural changes of myocardial cells in each group were observed under electron microscope. Results comparison between different treatment groups: morphological observation: the ultrastructure of cardiomyocytes in the blank control group was not changed under electron microscope. The ultrastructure of cardiomyocytes in oestradiol group, progesterone group, estradiol plus progesterone group and ovariectomized group were different. The ultrastructural changes of estradiol group, progesterone group and estradiol plus progesterone group were between control group and ovariectomized group. The statistical results of short-term treatment group and long-term treatment group showed that there were significant differences between the two groups in comparison with the other four groups. There was no significant difference between the OVX EB.HPC group and the INT group and the OVX HPC group, the OVX EB.HPC group and the OVX group, and the OVX HPC group and the OVX EB.HPC group were not significantly different from the other four groups. The difference between the two groups was statistically significant (P 0.01). OVX HPC group and OVX HPC group were significantly different from those in EB group.
