【摘要】:樹突狀細(xì)胞(dendritic cells,DCs)是體內(nèi)功能最強(qiáng)的抗原遞呈細(xì)胞。在過去的十年中,多數(shù)研究者的興趣是集中在DC是如何激發(fā)免疫應(yīng)答的研究上,然而,近幾年的工作顯示DCs不但具有激發(fā)免疫反應(yīng)的能力,而且,還可以下調(diào)免疫反應(yīng)或者誘導(dǎo)免疫耐受的產(chǎn)生,F(xiàn)在普遍認(rèn)為,DC在激活na(?)ve T細(xì)胞和誘導(dǎo)耐受中都扮演關(guān)鍵的角色。DCs既可以誘導(dǎo)中樞耐受又可以誘導(dǎo)外周耐受,特別是DCs在誘導(dǎo)外周耐受中的作用越來越受到重視。最近,具有免疫負(fù)向調(diào)節(jié)功能的調(diào)節(jié)性DCs(regulatory dendritic cells)被陸續(xù)發(fā)現(xiàn),并成為這一研究領(lǐng)域的新熱點(diǎn)。 目前對調(diào)節(jié)性DCs的研究主要集中在對其起源和發(fā)育的研究上,所采用的方法多數(shù)是用免疫抑制性細(xì)胞因子誘導(dǎo)產(chǎn)生的,如在常規(guī)誘導(dǎo)DCs的培養(yǎng)基中加入IL-10或TGF-β。但這類方法過于簡化,不能反映在體的實(shí)際分化過程,因此產(chǎn)生的DCs的特性是否能代表一種天然的細(xì)胞群體并不十分明確。 造血細(xì)胞的體內(nèi)發(fā)育過程是在一定的微環(huán)境的作用下進(jìn)行的。所謂的微環(huán)境(microenvironment)或“龕位”(niche)是由成纖維細(xì)胞、巨噬細(xì)胞、內(nèi)皮細(xì)胞、脂肪細(xì)胞等基質(zhì)細(xì)胞和它們分泌的細(xì)胞外基質(zhì)組成的,為造血細(xì)胞的發(fā)育提供各種必需的外源性信號的發(fā)育場所。不同的器官有不同的微環(huán)境,同一器官的不同區(qū)域的亞微環(huán)境也不同。不同的微環(huán)境可誘導(dǎo)產(chǎn)生不同的種類或功能特征的細(xì)胞。如骨髓的造血微環(huán)境。因此,脫離微環(huán)境來研究造血細(xì)胞的發(fā)育所得結(jié)論不可能反映機(jī)體的真實(shí)情況。 脾臟是機(jī)體最大的外周免疫器官,又是哺乳動(dòng)物的造血器官,并且小鼠脾臟終身保持造血功能。經(jīng)典的脾結(jié)節(jié)形成實(shí)驗(yàn)證明,脾臟的微環(huán)境不但具有支持造血的作用,而
[Abstract]:Dendritic cells ( DCs ) are the most potent antigen - presenting cells in vivo . In the past decade , the interest of most researchers has been focused on how DCs can stimulate immune responses . However , recent studies have shown that DCs play a key role in activating na ( ? ) ve T cells and inducing tolerance . It is generally believed that DCs play a key role in activating na ( ? ) ve T cells and inducing tolerance . DCs can both induce central tolerance and induce peripheral tolerance , especially DCs in inducing peripheral tolerance .
