牛奶中氨基糖苷類藥物殘留膠體金免疫層析技術(shù)研究

【摘要】：為建立可同時(shí)檢測(cè)牛奶中6種氨基糖苷類藥物殘留的膠體金免疫層析技術(shù),本研究采用棋盤法篩選膠體金顆粒標(biāo)記鏈霉素、慶大霉素及卡那霉素抗體的最佳pH值和最適抗體用量,將3種標(biāo)記好的金標(biāo)抗體按一定的比例混合凍干。實(shí)驗(yàn)結(jié)果表明:該多殘留膠體金免疫試紙條對(duì)牛奶中鏈霉素、雙氫鏈霉素、慶大霉素、小諾霉素、卡那霉素和妥布霉素的cut-off值分別為50、50、20、20、50、50 ng/m L。牛奶樣本無需前處理,同步檢測(cè)這6種藥物的時(shí)間在10 min以內(nèi)。采用膠體金方法檢測(cè)100份從全國(guó)不同區(qū)域采集的原奶樣品,并用高效液相色譜(HPLC)方法進(jìn)行鑒定,兩種方法的檢測(cè)結(jié)果基本一致,說明建立的膠體金免疫層析方法準(zhǔn)確性高,可靠性好。
[Abstract]:To establish a colloidal gold immunochromatography technique for simultaneous detection of 6 aminoglycoside residues in milk, a chessboard method was used to screen the best pH value and the optimum antibody dosage of colloidal gold particles labeled streptomycin, gentamicin and kanamycin antibody. The results of 3 kinds of gold labeled antibodies were mixed in a certain proportion to freeze dry. The results showed that the cut-off value of the multi residue colloid gold immunoassay paper for streptomycin, dihydrostreptomycin, gentamicin, novomycin, kanamycin and tobramycin was 50,50,20,20,50,50 ng/m L. milk samples without pre treatment, and the time of the 6 drugs was within 10 min synchronously. 100 copies were detected by colloidal gold method. The raw milk samples collected from different regions of China were identified by high performance liquid chromatography (HPLC). The results of the two methods were basically the same. The results showed that the colloidal gold immunochromatography (GI) was highly accurate and reliable.
