【摘要】：目的:探討幽門(mén)螺桿菌(Helicobacter pylori,H.pylori)作用巨噬細(xì)胞后導(dǎo)致的細(xì)胞炎癥反應(yīng)機(jī)制及對(duì)宿主巨噬細(xì)胞的損傷作用。方法:ELISA方法檢測(cè)幽門(mén)螺桿菌感染巨噬細(xì)胞后細(xì)胞培養(yǎng)上清液中細(xì)胞因子IL-23、IL-10、TNF-α及IL-8的含量,Western blot分析幽門(mén)螺桿菌作用巨噬細(xì)胞后細(xì)胞胞內(nèi)蛋白NOS2、COX2的表達(dá)水平;同時(shí)流式細(xì)胞技術(shù)分析幽門(mén)螺桿菌處理巨噬細(xì)胞后對(duì)細(xì)胞的凋亡作用。結(jié)果:細(xì)胞因子IL-23、IL-10、TNF-α、IL-8在幽門(mén)螺桿菌作用巨噬細(xì)胞后細(xì)胞培養(yǎng)上清中分泌顯著增多(P0.05),且NOS2、COX2蛋白表達(dá)顯著增強(qiáng)(P0.03);幽門(mén)螺桿菌處理巨噬細(xì)胞后巨噬細(xì)胞凋亡顯著增多(P0.05)。結(jié)論:幽門(mén)螺桿菌作用巨噬細(xì)胞后誘導(dǎo)巨噬細(xì)胞增強(qiáng)炎癥反應(yīng)而抑制或殺傷幽門(mén)螺桿菌,同時(shí)長(zhǎng)時(shí)間作用能誘導(dǎo)宿主巨噬細(xì)胞凋亡。
[Abstract]:Aim: to investigate the mechanism of inflammatory reaction induced by Helicobacter pylori (Helicobacter pylori,H.pylori) on macrophages and its damage to host macrophages. Methods: the levels of cytokine IL-23,IL-10,TNF- 偽 and IL-8 in the supernatant of Helicobacter pylori infected macrophages were detected by ELISA method., Western blot analysis of intracellular protein NOS2, after Helicobacter pylori infection on macrophages was carried out. The expression level of COX2; At the same time, flow cytometry was used to analyze the apoptosis of macrophages treated with Helicobacter pylori. Results: the levels of cytokines IL-23,IL-10,TNF- 偽 and IL-8 increased significantly in the supernatant of macrophages after Helicobacter pylori treatment (P0.05), and the expression of NOS2,COX2 protein increased significantly (P0.03). After treated with Helicobacter pylori, macrophages apoptosis increased significantly (P0.05). Conclusion: Helicobacter pylori can induce macrophages to enhance inflammatory response and inhibit or kill Helicobacter pylori.
