IL-17A-OVA疫苗對(duì)實(shí)驗(yàn)性自身免疫性葡萄膜炎的影響

【摘要】:目的研究IL-17A-OVA多肽疫苗對(duì)于實(shí)驗(yàn)性自身免疫性葡萄膜炎(EAU)的影響。設(shè)計(jì)實(shí)驗(yàn)研究。研究對(duì)象30只C57BL/6小鼠。方法制備IL-17A-OVA疫苗。將30只C57BL/6小鼠隨機(jī)分為疫苗組、佐劑組及環(huán)孢素組3組,每組10只,佐劑組及疫苗組提前免疫小鼠,第10周時(shí)三組小鼠同時(shí)用光感受器間維生素A類結(jié)合蛋白(IRBP)誘發(fā)EAU動(dòng)物模型。環(huán)孢素組造模成功后環(huán)孢素連續(xù)灌胃2周。HE染色觀察小鼠眼球活組織切片,ELISA方法檢測小鼠血清中IL-17A、IL-17A特異性抗體、IL-6及IL-23水平。主要指標(biāo)小鼠血清中IL-17A、IL-17A特異性抗體、IL-6及IL-23水平。結(jié)果組織病理學(xué)顯示疫苗組、環(huán)孢素組與佐劑組組織病理學(xué)評(píng)分均有顯著性差異(佐劑組評(píng)分較高),疫苗組與環(huán)孢素組組織病理學(xué)評(píng)分無顯著性差異;血清中IL-17A水平疫苗組(25.785±4.677 pg/ml)與環(huán)孢素組(23.129±3.940 pg/ml)均較佐劑組(33.774±6.478 pg/ml)顯著降低(F=8.125,P=0.003);IL-17A特異性抗體水平疫苗組(106.379±12.603 pg/ml)較環(huán)孢素組(59.431±8.626 pg/ml)及佐劑組(56.712±4.214pg/ml)顯著升高(F=63.008,P=0.000),環(huán)孢素組與佐劑組無顯著性差異;血清中IL-6水平疫苗組(41.124±4.959 pg/ml)與環(huán)孢素組(29.418±3.34 7 pg/ml)均較佐劑組(49.329±8.768 pg/ml)顯著降低(F=18.603,P=0.000);IL-23水平環(huán)孢素組(73.492±12.324 pg/ml)較佐劑組(109.770±20.848 pg/ml)及疫苗組(100.893±13.467 pg/ml)顯著降低,疫苗組與佐劑組無顯著性差異(F=4.916,P=0.018)。結(jié)論 IL-17A-OVA疫苗可通過產(chǎn)生IL-17A特異性抗體在減輕EAU的炎性反應(yīng)中起到一定的作用,該疫苗可能成為自身免疫性葡萄膜炎治療的新方法。
[Abstract]:Objective to study the effect of IL-17A-OVA polypeptide vaccine on (EAU) in experimental autoimmune uveitis. Design experimental research. Participants 30 C57BL/6 mice were studied. Methods IL-17A-OVA vaccine was prepared. Thirty C57BL/6 mice were randomly divided into three groups: vaccine group, adjuvant group and cyclosporine group with 10 mice in each group. The mice in adjuvant group and vaccine group were immunized in advance. At the 10th week, the three groups of mice were treated with (IRBP), a binding protein of vitamin A between photoreceptors, to induce EAU animal model at the same time. Cyclosporine was perfused intragastrically for 2 weeks after the model was established in cyclosporine group. HE staining was used to observe the living tissue sections of the eyeball of mice. The levels of IL-17A,IL-17A specific antibody, IL-6 and IL-23 in serum were detected by ELISA method. Main outcome measures IL-17A,IL-17A specific antibody, IL-6 and IL-23 levels in serum of mice. Results the histopathological scores of vaccine group, cyclosporine group and adjuvant group were significantly different (adjuvant group was higher), but there was no significant difference between vaccine group and cyclosporine group. The serum IL-17A levels in the vaccine group (25.785 鹵4.677 pg/ml) and the cyclosporine group (23.129 鹵3.940 pg/ml) were significantly lower than those in the adjuvant group (33.774 鹵6.478 pg/ml). IL-17A specific antibody level in vaccine group (106.379 鹵12.603 pg/ml) was significantly higher than that in cyclosporine group (59.431 鹵8.626 pg/ml) and adjuvant group (56.712 鹵4.214pg/ml). There was no significant difference between cyclosporine group and adjuvant group. The serum IL-6 levels in the vaccine group (41.124 鹵4.959 pg/ml) and the cyclosporine group (29.418 鹵3.347 pg/ml) were significantly lower than those in the adjuvant group (49.329 鹵8.768 pg/ml). The level of IL-23 in cyclosporine group (73.492 鹵12.324 pg/ml) was significantly lower than that in adjuvant group (109.770 鹵20.848 pg/ml) and vaccine group (100.893 鹵13.467 pg/ml). Conclusion IL-17A-OVA vaccine may play a role in alleviating the inflammatory response of EAU by producing IL-17A specific antibodies, and this vaccine may be a new method for the treatment of autoimmune uveitis.
