WEE1在腎透明細(xì)胞癌中的表達(dá)及意義

【摘要】:目的:研究W E E1基因在腎透明細(xì)胞癌(RCCC)中的表達(dá)和意義,分析W E E1基因與腎透明細(xì)胞癌的臨床分期、病理分級及淋巴轉(zhuǎn)移的關(guān)系,為腎細(xì)胞癌(主要為透明細(xì)胞癌)的診斷治療及預(yù)后提供理論指導(dǎo)。 方法:應(yīng)用免疫組織化學(xué)、real time PCR和Western blot法檢測32例腎透明細(xì)胞癌和25例癌旁正常腎組織中W E E1的表達(dá)情況,并將W E E1轉(zhuǎn)錄及表達(dá)水平與RCCC的生物學(xué)效應(yīng)進行相關(guān)分析;采用SPSS14.0軟件統(tǒng)計學(xué)分析實驗數(shù)據(jù),設(shè)定P0.05為差異具有統(tǒng)計學(xué)意義。 結(jié)果:32例腎透明細(xì)胞癌組織中W E E1的陽性表達(dá)為28例(87.5%),陰性表達(dá)為4例(12.5%),25例正常腎組織中W E E1陽性表達(dá)為2例(8.0%),陰性表達(dá)為23例(92%)。兩組間比較,差異具有統(tǒng)計學(xué)意義(P0.01);腎透明細(xì)胞癌組織和正常組織W E E1表達(dá)CT值分別為13.7±3.7和4.5±2.0,兩組比較有統(tǒng)計學(xué)差異(P0.01),腎癌組織的W E E l表達(dá)顯著大于正常組織中的W E E1表達(dá)。Western Blot檢測結(jié)果顯示,腎癌組織和正常組織W E E1表達(dá)相對灰度比值分別為5.2±1.5和0.6±0.37,兩組比較有統(tǒng)計學(xué)差異(P0.01),腎癌組織的W E E1表達(dá)顯著大于正常組織中的W E E1蛋白表達(dá)。W E E1表達(dá)水平與臨床分期密切相關(guān),Ⅰ、Ⅱ和Ⅲ-Ⅳ期相對灰度比值分別為3.1±0.7,4.7±0.9和6.7±0.9,其Ⅱ和Ⅲ-Ⅳ期腎癌分別是Ⅰ期的1.5倍和2.2倍。(P0.05)。W EE1表達(dá)水平與組織分級密切相關(guān),高分化、中分化和低分化腎癌灰度比值分別為3.1±0.5,4.4±0.5和6.4±0.9,中分化、低分化腎癌灰度比值分別是高分化腎癌的1.75倍、1.80倍(P0.05)。W E E1表達(dá)水平與淋巴轉(zhuǎn)移也密切相關(guān),淋巴無轉(zhuǎn)移組和轉(zhuǎn)移組W E E1灰度比值分別為4.3±1.1和6.8±0.6,淋巴轉(zhuǎn)移組W E E1灰度比值是無轉(zhuǎn)移組腎癌的1.6倍(P0.05)。W E E l表達(dá)和淋巴轉(zhuǎn)移呈正相關(guān),相關(guān)系數(shù)為0.805(P0.01)。 結(jié)論:1、W E E1在腎透明細(xì)胞癌轉(zhuǎn)錄與表達(dá)明顯高于對照組,對照組中W E E1低表達(dá)。2、W E E1的表達(dá)可能與腎透明細(xì)胞癌的臨床分期、病理分級、淋巴轉(zhuǎn)移相關(guān)。當(dāng)患者處于Ⅲ-Ⅳ期時,WEE1腎細(xì)胞癌中的表達(dá)量比處于Ⅰ、Ⅱ期患者腎細(xì)胞癌中表達(dá)高,中、低分化中的表達(dá)顯著大于高分化,在有淋巴轉(zhuǎn)移的腎透明細(xì)胞癌中的表達(dá)明顯高于無淋巴轉(zhuǎn)移的腎透明細(xì)胞癌。3、W E E1有望成為判斷腎透明細(xì)胞癌轉(zhuǎn)移和預(yù)后等生物學(xué)行為的重要標(biāo)記物,將有可能成為腎透明細(xì)胞癌治療的新靶點。
[Abstract]:Objective: to study the expression and significance of WEE1 gene in (RCCC) of renal clear cell carcinoma (RCC), and to analyze the relationship between WE1 gene and clinical stage, pathological grade and lymphatic metastasis of renal clear cell carcinoma (RCC). To provide theoretical guidance for the diagnosis, treatment and prognosis of renal cell carcinoma (mainly clear cell carcinoma). Methods: the expression of WE1 in 32 cases of renal clear cell carcinoma and 25 cases of adjacent normal renal tissues were detected by immunohistochemistry real time PCR and Western blot, and the correlation between the transcription and expression of WE1 and the biological effect of RCCC was analyzed. SPSS14.0 software statistical analysis of experimental data, set P0.05 as the difference has statistical significance. Results in 32 cases of renal clear cell carcinoma, 28 cases (87.5%) were positive for WE1, 4 (12.5%) for negative expression, 2 (8.0%) for WE1 and 23 (92%) for negative expression in 25 normal renal tissues. The difference between the two groups was statistically significant (P0.01). The CT expression of WEE1 in renal clear cell carcinoma and normal tissues were 13.7 鹵3.7 and 4.5 鹵2.0, respectively. There was a significant difference between the two groups (P0.01). The expression of W E E l in renal cell carcinoma was significantly higher than that in normal tissues.
