【摘要】:雞(Gallus gallus)和鴨(Anas platyrhynchos)是新城疫病毒(Newcastle disease virus,NDV)的主要宿主,但對于NDV感染的反應(yīng)差異顯著,先天性免疫應(yīng)答在其中起重要作用。為了比較鴨源NDV激活雞和鴨先天性免疫應(yīng)答的不同,闡明先天性免疫應(yīng)答在宿主致病性差異產(chǎn)生中的作用,以SD03(鴨源NDV)對雞和鴨的致病性比較研究為基礎(chǔ),利用熒光定量PCR對不同時間點外周血中模式識別受體(pattern recognition receptors,PRRs)、細胞因子、抗病毒蛋白和主要組織相容性復(fù)合體分子(major histocompatibility complex,MHC)m RNA的表達量進行檢測和比較。結(jié)果表明,SD03在雞脾臟中的復(fù)制效率高于鴨;免疫基因中Toll樣受體7(toll-like receptor 7,TLR-7)、白介素1β(interleukin 1β,IL-1β)、IL-2、IL-6、IL-8、干擾素β(interferonβ,IFN-β)、MHCⅠ和MHCⅡ在雞和鴨外周血中的表達模式不同且表達量變化趨勢差異顯著;除IL-2外,所有檢測基因在雞外周血中的表達量明顯高于鴨。研究結(jié)果在機體水平上展示了NDV誘導(dǎo)雞和鴨先天性免疫反應(yīng)的不同,并分析了其在致病性差異產(chǎn)生中的作用,為全面理解NDV對不同禽類的致病機制提供了數(shù)據(jù)支持。
[Abstract]:Gallus gallus) and Anas platyrhynchos) are the main hosts of Newcastle disease virus (NDV), but there are significant differences in the response to NDV infection, and innate immune response plays an important role. To elucidate the role of congenital immune response in the pathogenicity difference of host, based on the comparative study of pathogenicity between chicken and duck by SD03 (duck NDV), fluorescence quantitative PCR was used to detect the pattern recognition receptor pattern recognition receptor (PRRsN), cytokines in peripheral blood at different time points. The expression of MHCm RNA of major histocompatibility complex was detected and compared with that of major histocompatibility complex. The results showed that the replication efficiency of MHCM RNA in chicken spleen was higher than that in duck.
