細(xì)胞凋亡和細(xì)胞自噬在慢性環(huán)孢素A腎毒性中的作用

【摘要】:目的探討細(xì)胞凋亡和細(xì)胞自噬在慢性環(huán)孢素A(CsA)腎毒性中的作用。方法 Sprague-Dawley大鼠分為兩組:對照組:皮下注射橄欖油(1 ml·kg-1·d-1)4 w;慢性CsA腎毒性組:皮下注射CsA(15 mg·kg-1·d-1)4 w。檢測兩組大鼠的體重和腎功能;三色染色檢測腎小管間質(zhì)纖維化程度;ELISA法檢測血、尿8羥基脫氧鳥苷(8-OHdG)水平;原位末端標(biāo)記法(TUNEL)染色觀察細(xì)胞凋亡;免疫印跡法檢測細(xì)胞凋亡調(diào)控基因(Bcl-2、Bax、Caspase-3)和細(xì)胞自噬體膜型LC3-Ⅱ蛋白的表達(dá);Pearson直線相關(guān)分析腎小管間質(zhì)纖維化程度與細(xì)胞凋亡和LC3-Ⅱ蛋白表達(dá)的相關(guān)關(guān)系。結(jié)果與對照組相比,毒性組大鼠體重減輕、腎功能低下、血尿8-OHdG水平升高,同時腎毒性組可見明顯的腎小管間質(zhì)纖維化和大量TUNEL陽性細(xì)胞。免疫印跡結(jié)果表明,毒性組Bax、Caspase-3、LC3-Ⅱ蛋白的表達(dá)明顯增加,反之,Bcl-2的表達(dá)顯著減少。直線相關(guān)分析提示,腎小管間質(zhì)纖維化程度與TUNEL陽性細(xì)胞數(shù)和LC3-Ⅱ蛋白表達(dá)呈正向相關(guān)。結(jié)論細(xì)胞凋亡和細(xì)胞自噬參與了慢性CsA腎毒性腎小管間質(zhì)的損傷。
[Abstract]:Objective to investigate the role of apoptosis and autophagy in chronic cyclosporine (CSA) nephrotoxicity. Methods Sprague-Dawley rats were divided into two groups: the control group was subcutaneously injected with 1 ml kg-1 d ~ (-1) of olive oil for 4 weeks, and the chronic CSA nephrotoxicity group was subcutaneously injected with CSA 15 mg kg-1 d ~ (-1) for 4 weeks. The weight and renal function of the rats in the two groups were measured, the degree of renal tubulointerstitial fibrosis was detected by trichromatic staining and the levels of blood and urine 8-OHdG were detected by Elisa, apoptosis was observed by in situ end labeling and Tunel staining.
