霍亂弧菌腸毒素B亞單位基因在雙歧桿菌中克隆及表達(dá)的初步研究

【摘要】:研究背景: 霍亂(Cholera)是一種由霍亂弧菌(Vibrio cholerae)引起的烈性傳染病,一直以來都是各國傳染病防治的重點(diǎn),免疫接種是預(yù)防霍亂的最有效措施。以前的注射用疫苗不同程度存在著各種缺點(diǎn),研究也顯示針對霍亂這種腸道傳染病的疫苗,通過口服途徑給予的效果優(yōu)于其他任何胃腸外給予途徑,因?yàn)槟c道粘膜免疫在霍亂的免疫預(yù)防中發(fā)揮著主要作用。因此,開發(fā)安全、方便、價(jià)廉的口服疫苗成為霍亂免疫預(yù)防研究的熱點(diǎn),而以霍亂弧菌腸毒素(Cholera toxin,CT)為基礎(chǔ)的基因重組口服疫苗成為霍亂疫苗開發(fā)的主要方向。CT是霍亂弧菌主要的致病因子,它是分子量為84KD的腸毒素,由1個(gè)A亞單位(Cholera toxin Asubunit,CTA)和5個(gè)B亞單位(Cholera toxin B subunit,CTB)形成的五聚體共價(jià)連接而成,CTA為毒性亞單位,CTB為無毒的受體結(jié)合亞單位,后者是霍亂的主要保護(hù)性抗原?诜﨏TB可以刺激機(jī)體產(chǎn)生保護(hù)性粘膜抗體和血清抗體,發(fā)揮霍亂防治作用。此外,CTB還是一種很好的粘膜免疫佐劑,用于強(qiáng)化多糖、小肽等半抗原的免疫原性,與其他不相關(guān)抗原共同使用時(shí),可以促進(jìn)誘導(dǎo)其他抗原的粘膜免疫反應(yīng),,用于更廣泛的人類疫苗的發(fā)展。所以,為充分發(fā)揮CTB的免疫特性作用,探索CTB在各種宿主表達(dá)的研究日益增多,如細(xì)菌、植物、
[Abstract]:Research background:
Cholera (Cholera) is a kind of Vibrio cholerae (Vibrio cholerae) caused by infectious diseases, has been the focus of the prevention and treatment of infectious diseases, vaccination is the most effective measure for the prevention of cholera. Previous vaccines had many defects in different degrees. This study also showed that for cholera intestinal infectious disease vaccine that effect was given via the oral route is better than any other way for parenteral administration, intestinal mucosal immunity plays a major role in the immune prevention of cholera.
