【摘要】:幽門螺桿菌(Helicobacter pylori,H. pylori)作為唯一能在人體胃內(nèi)酸性環(huán)境中定植并長(zhǎng)期生存的致病菌,其生長(zhǎng)在體內(nèi)必然會(huì)受到各種不利條件的影響,H. pylori通過(guò)啟動(dòng)一系列的適應(yīng)性調(diào)節(jié)機(jī)制來(lái)抵抗這些脅迫因素以達(dá)到保種生存并導(dǎo)致疾病的目的。形態(tài)學(xué)變化是H. pylori的適應(yīng)性調(diào)控機(jī)制之一,在不同的脅迫條件下H. pylori可發(fā)生不同的形態(tài)學(xué)變化,蛋白質(zhì)是生命活動(dòng)的最終執(zhí)行者,無(wú)論何種表征蛋白質(zhì)在其中將起重要作用,比較蛋白質(zhì)組學(xué)是研究病原菌在各種脅迫因素下蛋白質(zhì)表達(dá)差異的理想工具,現(xiàn)今較普遍使用的技術(shù)體系包括雙向凝膠電泳,質(zhì)譜分析及生物信息學(xué)分析。膽汁是機(jī)體重要的天然抗病原菌物質(zhì),研究H. pylori在膽汁中的存活能力,對(duì)于揭示H. pylori感染與十二指腸潰瘍的發(fā)生規(guī)律有重要意義,為此我們研究了H. pylori在膽汁脅迫下的蛋白表達(dá)狀態(tài)。絲狀體是H. pylori除螺桿狀和球形體以外的一種成活形式,對(duì)絲狀體蛋白表達(dá)譜的研究,有助于揭示其發(fā)生規(guī)律。我們?cè)l(fā)現(xiàn)鞭毛絲帽蛋白與H. pylori結(jié)構(gòu)性群體結(jié)構(gòu)形成密切相關(guān),然而,對(duì)生物質(zhì)譜篩選的候選蛋白尚需作進(jìn)一步的功能驗(yàn)證,為此,我們構(gòu)建了H. pylori結(jié)構(gòu)性群體形成相關(guān)蛋白鞭毛絲帽蛋白基因缺失突變株,為進(jìn)一步驗(yàn)證此蛋白的功能及闡明H. pylori結(jié)構(gòu)性群體的形成機(jī)制奠定工作基礎(chǔ)。主要研究?jī)?nèi)容及研究結(jié)果如下: 一、膽汁對(duì)幽門螺桿菌的抑制及其蛋白質(zhì)表達(dá)譜的初步研究 膽汁脅迫是人體內(nèi)抑制H. pylori生長(zhǎng)的不利條件之一,許多研究也表明膽汁能夠抑制H. pylori的生長(zhǎng),但H. pylori仍能夠在膽汁豐富的十二指腸球部存在,而對(duì)其他肝膽系統(tǒng)的感染較少見(jiàn),因此我們推測(cè)膽汁的酸化影響其對(duì)H. pylori的抑制活性。為了研究H. pylori對(duì)膽汁脅迫的適應(yīng)性調(diào)
[Abstract]:Helicobacter pylori Helicobacter pylori H.pylori is the only bacterium that can colonize and survive in an acidic environment in the stomach of a human body. Its growth in vivo is bound to be affected by various adverse conditions. H. pylori protects against these stress factors by initiating a series of adaptive regulatory mechanisms to preserve the survival of species and lead to disease. Morphological changes are H. pylori. One of the adaptive regulatory mechanisms, Under different stress conditions, the morphological changes of H. pylori are different. Protein is the final executor of life activity, no matter what characterization protein will play an important role in it. Comparative proteomics is an ideal tool for studying the differences in protein expression of pathogens under various stress factors. Bile is an important natural antipathogenic substance in the body. It is important to study the survival ability of H. pylori in bile for revealing the regularity of H. pylori infection and duodenal ulcer. In this paper, we studied the protein expression of H. pylori under bile stress. The filamentous body is a survival form of H. pylori except the screw and spheroids, and the protein expression profile of the filamentous body is studied. We have found that flagellum cap protein is closely related to the formation of structural population structure of H. pylori. However, the candidate proteins screened by mass spectrometry need further functional verification. We constructed a mutant with deletion of flagellin gene associated with the structural colony formation of H. pylori. In order to further verify the function of the protein and elucidate the formation mechanism of the structural population of H. pylori, the main contents and results are as follows:. A preliminary study on the inhibition of bile on Helicobacter pylori and its protein expression profile. Bile stress is one of the unfavorable conditions for inhibiting the growth of H. pylori in human body. Many studies have also shown that bile can inhibit the growth of H. pylori, but H. pylori can still exist in the duodenal bulb where bile is abundant, but infection of other hepatobiliary systems is rare.
