含苯并噻唑骨架熒光探針的合成及離子識(shí)別研究

【摘要】:苯并噻唑類衍生物是重要的含氮雜環(huán)化合物。其應(yīng)用范圍涉及到生物學(xué),醫(yī)學(xué)等諸多領(lǐng)域,由于具有良好的熒光特性,使得人們不斷地對(duì)其進(jìn)行深入研究。本文以5-甲基-2-羥基-3-(2-苯并噻唑基)苯甲醛為底物設(shè)計(jì)合成了一系列的熒光探針,通過研究這些熒光探針的離子識(shí)別性能,得到了一些有意義的結(jié)果。具體內(nèi)容如下:1.設(shè)計(jì)合成了基于苯并噻唑的烯胺酮型熒光探針BTP并考察了其陰離子識(shí)別行為。在DMSO/H_2O(95/5)溶液中,BTP可通過比色和熒光兩種信號(hào)通道高選擇性識(shí)別CN-。當(dāng)向BTP溶液中加入CN-后,觀察到顏色由橙色變?yōu)辄S綠色,同時(shí)發(fā)生熒光增強(qiáng)并伴有藍(lán)移。BTP對(duì)CN-的識(shí)別過程具有極強(qiáng)的抗干擾能力,檢測(cè)限為3.94×10-7M。高分辨質(zhì)譜及核磁共振氫譜的研究結(jié)果表明,CN-的識(shí)別經(jīng)歷了其對(duì)烯胺酮BTP的親核加成過程。此外,通過簡(jiǎn)單的試紙實(shí)驗(yàn)證明了BTP對(duì)CN-檢測(cè)的實(shí)用性。2.設(shè)計(jì)合成了含苯并噻唑席夫堿類的探針L,L1,可以對(duì)不同的離子具有選擇性的識(shí)別。L能夠在乙醇溶液中實(shí)現(xiàn)對(duì)Zn~(2+)與Mg~(2+)的雙識(shí)別,且互不干擾,其結(jié)合比均為1:1。該探針具有選擇性好,響應(yīng)時(shí)間快等優(yōu)點(diǎn)。而傳感器L1,能夠在DMF/H_2O(1/1,v/v)的緩沖體系中識(shí)別ClO-,且L1為比率型熒光探針,與單一波長(zhǎng)發(fā)射的熒光探針相比,更能克服環(huán)境因素的干擾。3.設(shè)計(jì)合成了一種新的含苯并噻唑縮氨基硫脲類探針L2,L2能夠在PBS(10 mM,p H=7.4)中高選擇性的對(duì)Zn~(2+)進(jìn)行識(shí)別,且不受其他陽(yáng)離子的干擾。該探針具有較高的靈敏度,可以在十秒內(nèi)完成識(shí)別,具有較低的檢測(cè)限(1.42×10~(-6) M),這些結(jié)果表明探針L2在鋅離子的檢測(cè)方面具有潛在的應(yīng)用價(jià)值。試紙實(shí)驗(yàn)表明探針L2能夠很好地對(duì)環(huán)境中的鋅離子進(jìn)行實(shí)際檢測(cè)。4.設(shè)計(jì)合成了一種以2-(2′-羥基苯基)苯并噻唑?yàn)榈孜锏孽k觐愄结楲3,該探針可以在乙醇溶劑中對(duì)Al~(3+)進(jìn)行識(shí)別,且具有較高的選擇性。通過探針L3對(duì)Al~(3+)的滴定實(shí)驗(yàn),說明探針L3在5.5倍時(shí)達(dá)到飽和。另外,結(jié)合比實(shí)驗(yàn)表明,探針L3與Al~(3+)是通過1:1進(jìn)行結(jié)合的。
