過表達篩選發(fā)現(xiàn)果蠅miR-2參與調控線粒體穩(wěn)態(tài)(英文)

【目的】真核細胞內(nèi)線粒體穩(wěn)態(tài)由多條分子通路共同調控,其中miRNA的重要性在最近日益凸顯。但是對miRNA如何調控線粒體穩(wěn)態(tài)仍缺乏全面準確的認識。本研究旨在探究miRNA對線粒體穩(wěn)態(tài)的調控作用及機制。【方法】在黑腹果蠅Drosophila melanogaster幼蟲脂肪體細胞中對miRNA進行過表達篩選,監(jiān)測線粒體形態(tài)變化。通過生物信息學方法預測參與調控線粒體穩(wěn)態(tài)的miRNA下游靶標,并通過RNAi敲低實驗檢驗靶標基因對線粒體形態(tài)的影響!窘Y果】在黑腹果蠅幼蟲脂肪體中監(jiān)測了過表達106個miRNA對線粒體形態(tài)及果蠅發(fā)育的影響,發(fā)現(xiàn)21個miRNA過表達引起脂肪體發(fā)育異常,其中10個miRNA過表達可以導致幼蟲期致死,11個miRNA過表達導致蛹期致死。過表達miR-2導致黑腹果蠅脂肪體細胞中線粒體異樣聚集。序列分析表明miR-2很可能靶向調控Pink1基因。Pink1基因表達量確實因miR-2過表達而下調。遺傳互作實驗發(fā)現(xiàn)過表達Pink1基因可以拯救過表達miR-2引起的線粒體異樣聚集!窘Y論】結果說明miR-2很可能通過靶向Pink1基因參與調控線粒體穩(wěn)態(tài)。
