不同產(chǎn)地丹參莖葉UPLC指紋圖譜與化學(xué)模式識別研究

【摘要】:目的研究并建立不同產(chǎn)地丹參莖葉UPLC指紋圖譜,采用電噴霧離子源-四極桿-飛行時(shí)間質(zhì)譜(ESI-QTOF/MS)對其特征峰進(jìn)行定性研究,為丹參地上莖葉資源的綜合開發(fā)利用提供科學(xué)依據(jù)。方法采用UPLC法構(gòu)建丹參莖葉的指紋圖譜,色譜條件為ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7μm)色譜柱,以乙腈-0.1%甲酸水為流動相梯度洗脫,體積流量0.4 m L/min,檢測波長280 nm,柱溫35℃。質(zhì)譜檢測采用負(fù)離子模式;電壓3.0 k V;離子源溫度120℃;霧化溫度400℃;霧化氣800 L/h。同時(shí)應(yīng)用相似度評價(jià)、聚類分析和主成分分析(PCA)對實(shí)驗(yàn)數(shù)據(jù)進(jìn)行處理,以分析5個不同產(chǎn)地12批丹參莖葉樣品間的相似性及差異性,并通過UPLC/Q-TOF/MS對丹參莖葉共有峰進(jìn)行了定性分析。結(jié)果獲得丹參莖葉UPLC特征圖譜,共有峰13個。12批丹參莖葉的相似度為0.823~0.997,聚類分析、PCA結(jié)果與相似度分析結(jié)果一致。結(jié)論丹參莖葉UPLC指紋圖譜的構(gòu)建和化學(xué)模式的識別為其質(zhì)量控制和丹參非藥用部位利用價(jià)值的挖掘提供科學(xué)依據(jù)。
