【摘要】：應(yīng)用UPLC-Q-TOF-MS技術(shù)和血清藥物化學(xué)理論分析檢測(cè)芩百清肺濃縮丸的入血成分。灌胃給予大鼠芩百清肺濃縮丸溶液后,肝門靜脈采血,血清經(jīng)SPE處理,采用ACQUITY UPLC BEH C_(18)色譜柱(2.1 mm×100 mm,1.7μm),0.1%甲酸水(A)-0.1%甲酸乙腈(B)流動(dòng)相系統(tǒng)進(jìn)行梯度洗脫,流速0.3 m L·min~(-1),柱溫35℃。通過對(duì)比分析體外樣品、空白血清及含藥血清,同時(shí)結(jié)合Peakview和Metabolitepilot軟件鑒定和表征其血中移行成分,在給藥后的大鼠血清中分析鑒定了28個(gè)血中移行成分,其中18個(gè)原型成分,10個(gè)代謝產(chǎn)物。應(yīng)用UPLC-Q-TOF-MS技術(shù)和血清藥物化學(xué)理論較為全面地闡明了芩百清肺濃縮丸在大鼠血清中的移行成分,為進(jìn)一步研究其體內(nèi)代謝過程以及藥效物質(zhì)基礎(chǔ)提供了科學(xué)依據(jù)。
[Abstract]:The components of Qinbai Qingfei concentrated Pill were detected by UPLC-Q-TOF-MS technique and serum pharmacochemical theory. After the rats were given Qinbai Qingfei concentrated Pill solution by gavage, blood was collected from portal vein of liver and serum was treated with SPE. The gradient elution was carried out by ACQUITY UPLC BEH C _ (18) column (2.1 mm 脳 100 mm,1.7 渭 m) and 0.1% formic acid water (A) -0.1% acetonitrile (B) mobile phase system.
