UPLC-Q-TOF-MS法鑒定連翹脂素在人肝微粒體中的代謝產(chǎn)物

【摘要】:目的建立體外人源肝微粒體孵育體系,應(yīng)用UPLC-Q-TOF-MS法,首次對(duì)連翹脂素在人肝微粒體中的代謝產(chǎn)物進(jìn)行鑒定。方法采用Phenomenex Kinetex C_(18)色譜柱(2.1 mm×100 mm,2.6μm),以0.1%甲酸水-乙腈為流動(dòng)相進(jìn)行梯度洗脫,流速為400μL·min~(-1);進(jìn)樣量5μL。采用正離子模式進(jìn)行檢測(cè),電噴霧離子源,源溫度為550℃,質(zhì)量數(shù)掃描范圍m/z 100~1 000。結(jié)果對(duì)ESI-MS正離子模式下連翹脂素可能的裂解途徑進(jìn)行了推測(cè),同時(shí)鑒定了連翹脂素在肝微粒體中的8個(gè)代謝產(chǎn)物。結(jié)論所建立的連翹脂素在人肝微粒體中的UPLC-Q-TOF-MS測(cè)定法方便快捷,可應(yīng)用于連翹脂素在人肝微粒中的代謝研究,同時(shí)為連翹脂素的藥物代謝動(dòng)力學(xué)提供了依據(jù)。
