3種獐牙菜屬民族藥UPLC指紋圖譜研究

【摘要】:目的采用UPLC技術(shù)結(jié)合定量分析、系統(tǒng)聚類分析(HCA)和主成分分析(PCA),評價、鑒別3種民族藥用植物川東獐牙菜Swertia davidii、狹葉獐牙菜S.angustifolia和紫紅獐牙菜S.punicea Hemsl.。方法采集3種獐牙菜共27株樣品色譜圖,將色譜圖導入中藥色譜指紋圖譜相似度評價系統(tǒng)軟件,確定樣品保留時間和峰面積數(shù)據(jù),對3組樣品進行相似度分析。峰面積數(shù)據(jù)導入SPSS和SIMCA-P~+軟件進行HCA和PCA,從樹狀圖和主成分得分圖中判斷聚類效果。結(jié)果該方法線性關(guān)系良好(R~20.999 3,儀器精密度高(RSD為0.19%~2.45%),方法重現(xiàn)性好(RSD為0~1.77%),加樣回收率在95.8%~102.3%,RSD為0.45%~2.81%。不同物種獐牙菜植物中指示化合物的量差異大,同一物種不同個體中指示化合物量差異明顯。獐牙菜苦苷在川東獐牙菜中為0.54~10.05 mg/g,狹葉獐牙菜中未檢出,紫紅獐牙菜中為1.76~15.62 mg/g;龍膽苦苷在川東獐牙菜中為4.27~11.18 mg/g,狹葉獐牙菜中為0.26~3.58 mg/g,紫紅獐牙菜中為1.13~16.11 mg/g;當藥苷在川東獐牙菜中為0.02~0.28 mg/g,狹葉獐牙菜中為0.02~0.14 mg/g,紫紅獐牙菜中為0.11~44.52 mg/g;芒果苷在川東獐牙菜中為0.14~0.55 mg/g,狹葉獐牙菜中為0.03~0.04 mg/g,紫紅獐牙菜中為0.01~13.49 mg/g。HCA樹狀圖分類準確率為85.2%,誤判的紫紅獐牙菜樣品中化合物量較低。PCA得分圖可以區(qū)分3種獐牙菜,紫紅獐牙菜聚類效果分散,個體之間差異較大。結(jié)論采用UPLC技術(shù)分析指示化合物的量,結(jié)合化學計量學能夠評價和鑒別3種獐牙菜屬藥用植物。在本研究中,紫紅獐牙菜表現(xiàn)出更高的藥用價值。
[Abstract]:Objective To use the UPLC technique to combine quantitative analysis, systematic cluster analysis (HCA) and principal component analysis (PCA) to evaluate and identify the three ethnic medicinal plants, such as Swertia davidii, S. angustifolia and S. panicea Hemsl. Methods A total of 27 samples were collected from three kinds of dishes. The chromatogram was introduced to the similarity evaluation system software of the fingerprint of the traditional Chinese medicine. The retention time and peak area data of the samples were determined, and the similarity of the three samples was analyzed. Peak area data was imported into SPSS and SIMA-P ~ + software for HCA and PCA, and the clustering effect was judged from the tree view and the main component score map. Results The linear relationship of the method is good (R-20.999 3, the precision of the instrument is high (the RSD is 0.19%-2.45%), the reproducibility of the method is good (RSD is 0-1.77%), the recovery rate of the sample is 95.8%-102.3%, and the RSD is 0.45%-2.81%. The difference in the amount of the indicated compounds in the plants of the different species was large, and the amount of the compound indicated in the different individuals of the same species was significant. 0.54-10.05mg/ g, 0.54-10.05mg/ g, 1.76-15.62mg/ g, and 1.76-15.62mg/ g in the tarragmus viola, 0.26-3.58mg/ g in the tarragmus, and 1.13-16.11 mg/ g in the tarragmus violaceum, and 0.02-0.28 mg/ g in the tarragon in the eastern Sichuan province. 0.02-0.14 mg/ g, 0.11-44.52 mg/ g, and 0.03-0.04mg/ g, and 0.01-13.49 mg/ g. The classification accuracy of HCA tree-tree was 85.2%. The PCA score map can be used to distinguish three kinds of tarragon, and the clustering effect of the purple-red and red-rooted vegetables is different, and the difference between the individual and the individual is large. Conclusion UPLC technique is used to analyze the amount of the indicated compound, and the combination of chemometrics can be used to evaluate and identify three kinds of Chinese medicinal plants. In this study, the purple and red-tooth dish showed higher medicinal value.
