對(duì)氨基水楊酸鈉對(duì)錳致大鼠肝抗氧化酶和病理學(xué)改變的影響

【摘要】:目的探討對(duì)氨基水楊酸鈉(PAS-Na)對(duì)錳致大鼠肝微粒體、線粒體抗氧化酶及病理學(xué)改變的影響。方法50只雄性SD大鼠按體重隨機(jī)分為對(duì)照組和染錳組。染錳組腹腔注射(ip)Mn Cl2·4H2O 15 mg/kg,對(duì)照組ip等容量生理鹽水,1次/d,每周5 d,連續(xù)6周。隨后,將染錳組按體重隨機(jī)分為染錳、低、中、高劑量PAS-Na治療(L、M、H-PAS)組。L、M、H-PAS組大鼠分別背部皮下注射(sc)PAS-Na 100、200和300 mg/kg,對(duì)照組、染錳組背部sc等容量生理鹽水,1次/d,每周4 d,連續(xù)4周。測定肝線粒體、微粒體超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)活性,并觀察肝組織病理學(xué)改變。結(jié)果與對(duì)照組比較,染錳組肝線粒體SOD和GSH-Px、微粒體SOD活性降低,但差異無統(tǒng)計(jì)學(xué)意義(P0.05)。染錳組肝臟系數(shù)增大,肝臟匯管區(qū)出現(xiàn)炎性淋巴細(xì)胞及中性粒細(xì)胞浸潤,肝小葉內(nèi)出現(xiàn)點(diǎn)狀壞死,甚至部分灶性壞死,部分肝細(xì)胞出現(xiàn)脂肪變性;與染錳組比較,M-PAS組點(diǎn)狀壞死灶明顯減少,L、H-PAS組點(diǎn)狀壞死灶也有所減少。結(jié)論染錳可使大鼠肝臟匯管區(qū)出現(xiàn)炎性淋巴細(xì)胞及中性粒細(xì)胞浸潤,肝小葉點(diǎn)狀壞死,甚至部分灶性壞死,肝細(xì)胞脂肪變性,PAS-Na治療可使其病理形態(tài)學(xué)改變明顯好轉(zhuǎn)。
[Abstract]:Objective to investigate the effects of sodium p-aminosalicylate (PAS-Na) on hepatic microsomes, mitochondrial antioxidant enzymes and pathological changes in rats induced by manganese. Methods 50 male Sprague-Dawley rats were randomly divided into two groups according to their body weight: control group and manganese group. In the manganese exposed group, (ip) mn Cl 2 4H 2O was injected intraperitoneally at 15 mg / kg, while in the control group, the same volume of normal saline was given once a day, 5 days a week for 6 weeks. Then, according to the body weight, the rats in the manganese exposed group were randomly divided into three groups: manganese, low, medium and high dose PAS-Na treatment group. The rats were subcutaneously injected with (sc) PAS-Na 100200 and 300mg / kg in the back of the control group and the manganese exposed group respectively. The rats in the control group and the manganese exposed group received sc isovolumic physiological saline once a day, 4 days a week for 4 weeks. The activities of liver mitochondria, microsomal superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured. Results compared with the control group, the activities of SOD and GSH-Px in liver mitochondria and microsomal SOD in manganese exposed group were lower than those in control group, but there was no significant difference (P0.05). In the manganese exposed group, the liver coefficient increased, inflammatory lymphocytes and neutrophils infiltrated in the hepatic catchment area, dotted necrosis or even partial focal necrosis in the hepatic lobules, and fatty degeneration in some liver cells. Compared with manganese group, the point necrotic foci in M-PAS group were significantly reduced, and that in LH-PAS group was also decreased. Conclusion Manganese can make inflammatory lymphocytes and neutrophils infiltrate in the hepatic catchment area of rats, and hepatic lobular dotted necrosis, even partial focal necrosis. PAS-Na treatment of hepatocyte steatosis can obviously improve the pathomorphologic changes of rats.
