基于復(fù)制-轉(zhuǎn)錄碰撞的突變和進(jìn)化意義(英文)

復(fù)制和轉(zhuǎn)錄機(jī)器會(huì)同時(shí)使用相同的DNA區(qū)域作為模板,因此復(fù)制和轉(zhuǎn)錄不可避免地以頭對(duì)頭或追尾方式相互碰撞.頭對(duì)頭碰撞和追尾碰撞均會(huì)導(dǎo)致復(fù)制機(jī)器停留,從而造成DNA損傷和基因組不穩(wěn)定.就基因組完整性而言,頭對(duì)頭碰撞比追尾碰撞的后果更嚴(yán)重.本文回顧總結(jié)了復(fù)制-轉(zhuǎn)錄沖突的解決機(jī)制和進(jìn)化影響.相對(duì)于前導(dǎo)鏈,滯后鏈上非同義(氨基酸改變)突變的發(fā)生率更高,并且滯后鏈上基因的高頻誘變?nèi)Q于轉(zhuǎn)錄本和基因大小,因此較快的適應(yīng)性突變發(fā)生在滯后鏈上.頭對(duì)頭基因的高度轉(zhuǎn)錄增加了復(fù)制過程中響應(yīng)壓力的突變率.無論是頭對(duì)頭還是追尾模式,復(fù)制-轉(zhuǎn)錄沖突都可能是適應(yīng)性進(jìn)化的驅(qū)動(dòng)力.
