【摘要】:目的探討四川省成都地區(qū)發(fā)熱呼吸道住院患者人鼻病毒(HRV)感染的分子流行病學特征,為本地人鼻病毒感染的防控積累資料。方法于2014-2016年采集成人發(fā)熱呼吸道癥候群住院患者痰液1 040份,提取標本核酸,利用逆轉(zhuǎn)錄聚合酶鏈式反應(RT-PCR)以及巢式PCR(Nested-PCR)擴增HRV-5'NCR基因,篩查HRV陽性標本,隨機選取61份陽性標本擴增VP4/VP2基因,測定核酸序列并進行同源性比對和分型。結(jié)果 1 040例標本核酸檢出率為10.9%;冬春季檢出率高,隨年齡升高檢出率呈上升趨勢;獲得的61條VP4/VP2序列中,30條為HRV-A型,10條為HRV-B型,21條為HRV-C型;并發(fā)現(xiàn)HRV基因型內(nèi)和型間核酸同源性差異均較大。結(jié)論成都地區(qū)成人發(fā)熱呼吸道癥候群住院患者HRV的感染率高,以A型與C型流行為主,病毒VP4/VP2核酸片段變異較大。
[Abstract]:Objective to investigate the molecular epidemiological characteristics of human rhinovirus (HRV) infection in hospitalized patients with fever and respiratory tract in Chengdu, Sichuan Province. Methods from 2014 to 2016, 1 040 samples of sputum from adult patients with febrile respiratory syndrome were collected and their nucleic acids were extracted. The HRV-5'NCR gene was amplified by reverse transcriptase polymerase chain reaction (RT-PCR) and nested PCR-Nested-PCR. HRV positive samples were screened. VP4/VP2 gene was amplified from 61 positive samples randomly, nucleic acid sequence was determined, homologous alignment and typing were carried out. Results the detection rate of nucleic acid was 10.9% in 1 040 samples. The detection rate was high in winter and spring and increased with age. Among the 61 VP4/VP2 sequences, 30 were HRV-A and 10 were HRV-B. 21 were HRV-C. It was also found that the homology of nucleic acid in and between HRV genotypes was significant. Conclusion the infection rate of HRV in adult patients with febrile respiratory syndrome in Chengdu area is high, mainly in type A and type C. VP4/VP2 nucleic acid fragments of the virus vary greatly.
