兩基原中藥決明子UPLC指紋圖譜研究

【摘要】:目的建立兩基原(決明、小決明)中藥決明子的UPLC指紋圖譜,為決明子藥材質(zhì)量控制和評價提供依據(jù)。方法采用Agilent ZORBAX EP C18(100 mm×3.0 mm,1.8μm)色譜柱;以乙腈-0.01%甲酸水溶液為流動相,梯度洗脫,體積流量0.4 m L/min,檢測波長285 nm,柱溫30℃,進樣量為1μL。結(jié)果首次建立了決明子UPLC指紋圖譜共有模式,運用相似度評價軟件標(biāo)定了20個共有峰,結(jié)合保留時間和紫外光譜分析,指認了7個峰;20批決明子藥材相似度均大于0.9;聚類分析結(jié)果為當(dāng)歐式距離平方和為5時,20批決明子樣品可分為4類;主成分分析降維得到3個主成分,根據(jù)主成分得分將樣品進行劃分歸類,與聚類分析結(jié)果一致;通過擬合歸納第一主成分的載荷因子模型,篩選出紅鏈霉素-6-O-β-龍膽二糖苷等具有評價決明子質(zhì)量優(yōu)劣的特征峰。結(jié)論決明與小決明共有化學(xué)成分存在共性差異,該方法可以用于決明子藥材質(zhì)量控制和評價。
