MOF材料負(fù)載酶及其催化性能研究

【摘要】:金屬-有機(jī)骨架材料是一種近二十年來(lái)新興的納米多孔材料,結(jié)構(gòu)類似于沸石,具有較高的孔隙率與比表面積,已在能源氣體存儲(chǔ)、吸附分離、膜分離、多相催化、藥物控釋和光電磁等領(lǐng)域廣泛應(yīng)用。此外,將MOF材料用于酶固定,可以解決酶難以回收、無(wú)法重復(fù)使用等不足。酶固定指的是采用載體負(fù)載酶,例如吸附、包埋等方法,使酶限制于載體的表面以及微孔中,同時(shí)負(fù)載后的酶仍具有與游離酶類似的催化活性,以達(dá)到回收酶重復(fù)利用的目的。酶固定具有很多優(yōu)點(diǎn),如,固定化酶具有更好的穩(wěn)定性,易于儲(chǔ)存和運(yùn)輸,并且可以反復(fù)使用,實(shí)現(xiàn)重復(fù)利用。本工作通過(guò)實(shí)驗(yàn)的方法,采用了模板劑法溶劑熱合成多級(jí)孔UiO-66,與文獻(xiàn)發(fā)表的材料一致,BET為917 m2.g-1。進(jìn)一步用其負(fù)載了胃蛋白酶。負(fù)載方法為吸附法,在35℃,pH=3.6的負(fù)載條件下,多級(jí)孔UiO-66、PCN-222、SBA-15固定胃蛋白酶的最大吸附量分別為0.96 g/g、0.755 g/g、0.366 g/g.本工作以血紅蛋白作為底物,測(cè)定了胃蛋白酶和固定化酶的酶活,并且考察了固定化酶的重復(fù)利用性。測(cè)得濃度分別為1 mg/mL和0.8 mg/mL時(shí)的胃蛋白酶游離酶活分別為17.0×103U/g和18.5×103U/g.多級(jí)孔UiO-66吸附固定2 mg/mL胃蛋白酶的酶活為:1.1×103U/g.PCN-222和SBA-15固定后酶活較低。35℃下,在pH為3.6的醋酸緩沖液環(huán)境下進(jìn)行酶催化反應(yīng),1h后,過(guò)濾出固體,用醋酸緩沖液洗凈凍干。用上述方法測(cè)得酶活為0.65×103U/g,酶活回收率為56.5%.同樣條件下反應(yīng)2h,過(guò)濾洗凈凍干后,測(cè)得酶活為0.57×103U/g,酶活回收率為49.3%.
[Abstract]:The metal-organic skeleton material is a new kind of nano-porous material in recent 20 years. Its structure is similar to that of zeolite. It has high porosity and specific surface area. It has been used in energy gas storage, adsorption and separation, membrane separation, and heterogeneous catalysis. Drug controlled release and optoelectronic magnetism are widely used. In addition, the application of MOF material to enzyme fixation can solve the problems of enzyme recovery and reuse. Enzyme immobilization refers to the use of carrier loading enzymes, such as adsorption, entrapping, etc., so that the enzyme is confined to the surface and micropore of the carrier, while the enzyme after loading still has the same catalytic activity as the free enzyme, in order to achieve the purpose of reusing the enzyme. Enzyme fixation has many advantages, such as better stability, easy storage and transport, and can be reused. In this work, the method of templating was used to synthesize multiporous UiO-66, by solvothermal method, and the BET was 917m2.g-1m ~ (-1). It is further loaded with pepsin.
