含金屬卟啉的水溶性共軛聚電解質的設計、合成及其生物應用

【摘要】:水溶性共軛聚合物(WSCPs)通常由控制其主要光學性質的π共軛主鏈和控制其水溶性的側鏈組成。WSCPs在生物醫(yī)學方面具有很多優(yōu)勢,如高的亮度、大的消光系數(shù)、優(yōu)異的耐光性、低的細胞毒性和良好的水溶液穩(wěn)定性。金屬卟啉是生命存在的基石之一,在自然界中有獨特的生物學功能。它們都被廣泛的應用在生物傳感,熒光成像,醫(yī)學和光動力治療等領域。本文結合WSCPs和金屬卟啉兩者的優(yōu)勢,合成了一類含金屬卟啉的WSCPs材料,研究了其在細胞成像、磁共振成像(MRI)和光動力治療(PDT)方面的應用。1、通過將具有T1加權成像功能的錳卟啉引入到聚芴基共軛聚合物中,合成了具有熒光/MRI雙模式成像功能的水溶性共軛聚電解質。雙親性的化學結構使其可以在PBS緩沖溶液中經過自組裝形成100 nm左右粒徑均一的球形納米粒子,其光物理性質經過紫外-可見吸收光譜和熒光光譜進行了表征,形貌通過射電子掃描顯微鏡(TEM)和動態(tài)光散射(DLS)進行了表征。結果表明水溶性共軛聚合物納米粒子具有強的吸收、高的熒光量子產率、好的穩(wěn)定性和生物相容性、優(yōu)異的細胞成像和高的弛豫效率值(r1=13.15 mM-1S-1),這些結果表明其在活細胞成像和MRI方面有很好的應用前景。2、設計合成了一種含有金屬鈀卟啉的新型共軛聚合物,借助其兩親性的分子結構,在水溶液中經過自組裝形成30 nm左右的納米粒子。利用核磁(NMR)和凝膠滲透色譜(GPC)對中間體和目標聚合物的結構進行了表征。利用TEM和DLS對水溶性共軛聚合物納米粒子的形貌進行了研究。實驗結果表明該聚合物納米粒子粒徑均一,具有良好的分散性和穩(wěn)定性,可以進行細胞染色。重要的是,水溶性共軛聚合物納米粒子在光照條件下可以有效地產生單線態(tài)氧,使腫瘤細胞凋亡,實現(xiàn)了細胞成像和PDT的目的。
[Abstract]:The water-soluble conjugated polymer (WSCPs) is usually composed of 蟺 conjugated main chains which control its main optical properties and side chains that control its water solubility. WSCPs have many advantages in biomedical fields, such as high brightness, large extinction coefficient, excellent light resistance. Low cytotoxicity and good stability of aqueous solution. Metalloporphyrin is one of the cornerstones of life and has unique biological functions in nature. They are widely used in biosensor, fluorescence imaging, medicine and photodynamic therapy. Combined with the advantages of WSCPs and metalloporphyrin, a class of WSCPs materials containing metalloporphyrin were synthesized, and their cellular imaging was studied. The application of magnetic resonance imaging (MRI) and photodynamic therapy (PDT). 1. By introducing manganese porphyrin with T1 weighted imaging into polyfluorene conjugated polymer, a water-soluble conjugated polyelectrolyte with fluorescence / MRI dual-mode imaging was synthesized. The amphiphilic chemical structure made it possible to self-assemble in PBS buffer solution to form spherical nanoparticles with about 100 nm homogeneous diameter. Their photophysical properties were characterized by UV-Vis absorption and fluorescence spectra. The morphology was characterized by (TEM) and (DLS). The results show that the water-soluble conjugated polymer nanoparticles have strong absorption, high fluorescence quantum yield, good stability and biocompatibility. Excellent cell imaging and high relaxation efficiency (R1C 13.15 mM-1S-1). These results show that it has a good application prospect in living cell imaging and MRI. A novel conjugated polymer containing PD porphyrin was designed and synthesized. With the help of its amphiphilic molecular structure, nanoparticles were self-assembled in aqueous solution for about 30 nm. The structures of intermediates and target polymers were characterized by NMR (NMR) and gel permeation chromatography (GPC). The morphology of water-soluble conjugated polymer nanoparticles was studied by TEM and DLS. The results show that the polymer nanoparticles have uniform particle size, good dispersion and stability, and can be stained by cells. It is important that the water-soluble conjugated polymer nanoparticles can effectively produce singlet oxygen under illumination, which can make tumor cells apoptosis and achieve the purpose of cell imaging and PDT.
