超臨界流體色譜與高效液相色譜分離手性化合物的比較

【摘要】:超臨界流體色譜(SFC)分離具有速度快、分離效率高、溶劑消耗少等優(yōu)點(diǎn),近年來在手性化合物的分離分析中得到諸多應(yīng)用。本文對(duì)比研究了涂覆型多糖手性色譜柱在SFC和高效液相色譜(HPLC)上拆分24種手性化合物的差異。通過比較這些化合物在色譜柱上的保留時(shí)間和選擇因子等發(fā)現(xiàn)多數(shù)化合物在SFC上的分離效率要高于其在HPLC上的分離效率,但HPLC對(duì)軸手性化合物的分離效率要優(yōu)于SFC。SFC和HPLC的分離表現(xiàn)出一定的互補(bǔ)性,隨著苯環(huán)側(cè)鏈烷基的碳數(shù)增加,化合物在SFC上的保留逐漸增強(qiáng),而在HPLC的保留卻逐漸減弱。葉菌唑在使用SFC和HPLC分析時(shí)出現(xiàn)了洗脫順序反轉(zhuǎn)的現(xiàn)象。這些結(jié)果為SFC手性拆分提供了參考。
[Abstract]:Supercritical fluid chromatography (SFC) separation has many advantages, such as high speed, high separation efficiency and less solvent consumption. In recent years, it has been widely used in the separation and analysis of chiral compounds.In this paper, the separation of 24 chiral compounds on SFC and high performance liquid chromatography (HPLC) on coated polysaccharide chiral column was studied.By comparing the retention time and the selection factor of these compounds on the chromatographic column, it was found that the separation efficiency of most compounds on SFC was higher than that on HPLC.However, the separation efficiency of HPLC was better than that of SFC.SFC and HPLC. With the increase of the alkyl number of benzene ring, the retention of the compounds on SFC gradually increased, but the retention in HPLC gradually weakened.The reversal of elution sequence was observed in the use of SFC and HPLC analysis.These results provide a reference for SFC chiral resolution.
