【摘要】：目的:探討TLR4基因敲除對(duì)小鼠免疫細(xì)胞及脂肪因子的影響。方法:取20周齡的雄性野生型C57BL/6小鼠和TLR4~(-/-)小鼠的脾臟和附睪脂肪組織,分離細(xì)胞,用流式檢測(cè)F4/80、CD11b、CD11c、CD3、CD4、CD8分子的表達(dá);q PCR檢測(cè)附睪脂肪組織內(nèi)IL-6、HMGB1、TNF-α、脂聯(lián)素和抵抗素的表達(dá)。結(jié)果:與野生型C57BL/6小鼠相比,TLR4~(-/-)小鼠脾臟和附睪脂肪組織中M1型(F4/80~+CD11b~+CD11c~+)巨噬細(xì)胞比例上升(P0.05),M2型(F4/80~+CD11b~+CD11c~-)巨噬細(xì)胞比例下降(P0.05),這種趨勢(shì)在附睪脂肪組織中表現(xiàn)更為顯著。同時(shí)發(fā)現(xiàn)附睪脂肪組織中CD4~+T細(xì)胞比例下降(P0.05),CD8+T細(xì)胞比例上升(P0.05);IL-6、HMGB1、抵抗素表達(dá)升高(P0.05);TNF-α和脂聯(lián)素表達(dá)降低(P0.05)。結(jié)論:TLR4基因敲除可導(dǎo)致內(nèi)臟脂肪組織脂肪因子和免疫細(xì)胞的紊亂。
[Abstract]:Objective: to investigate the effect of TLR4 knockout on immune cells and fat factors in mice. Methods: the spleen and epididymal adipose tissues were isolated from male wild-type C57BL/6 mice and TLR4- / -r-mice at the age of 20 weeks. The expressions of IL-6HMGB1TNF- 偽, adiponectin and resistin in epididymal adipose tissues were detected by flow cytometry. Results: compared with wild-type C57BL/6 mice, the ratio of M _ 1 type F _ 4 / F _ 4 ~ CD _ 11b ~ CD _ 11c ~) macrophages in the spleen and epididymal adipose tissue of TLR4 / r ~ (-) mice was increased, and the percentage of M _ (1) F _ (4 / 80) ~ CD _ (11) C ~ (-) M _ (2) M _ (2) F _ (4 / 80 ~ CD _ (11) b ~ CD _ (11) C ~ (-) macrophages decreased significantly in epididymal adipose tissue, which was more significant in epididymal adipose tissue. At the same time, it was found that the percentage of CD4- T cells in adipose tissue of epididymis was decreased. The percentage of CD8 T cells in epididymal adipose tissue was increased, the expression of resistin was increased, the expression of adiponectin was decreased, and the expression of adiponectin was decreased. Conclusion the knockout of the TLR4 gene may lead to the disturbance of adipogenic factors and immune cells in visceral adipose tissue.
