鈣對生長期染鉛大鼠免疫損傷干預機制的研究

【摘要】:[目的]通過觀察不同劑量鈣對染鉛大鼠體內(nèi)二價金屬元素、白細胞數(shù)、血清IgG、脾臟指數(shù)和脾臟組織形態(tài),及免疫相關(guān)細胞因子基因的mRNA表達量等的影響,探索鈣對生長期染鉛大鼠免疫損傷的干預機制,進而為有效預防鉛中毒提供理論依據(jù)。[方法]選用21日齡清潔級SD雄性大鼠60只,適應性喂養(yǎng)7d后,按體重隨機分為5組,每組12只,分別為對照組(基礎(chǔ)飼料,去離子水)、染鉛組(基礎(chǔ)飼料,0.2%醋酸鉛水)、試驗Ⅰ組(基礎(chǔ)飼料添加0.5%碳酸鈣,0.2%醋酸鉛水)、試驗Ⅱ組(基礎(chǔ)飼料添加1.0%碳酸鈣,0.2%醋酸鉛水)、試驗Ⅲ組(基礎(chǔ)飼料添加2.0%碳酸鈣,0.2%醋酸鉛水)。飼養(yǎng)60d后,采集全血、肝臟、腎臟、脾臟、心臟、肌肉,測定其Pb、Ca、Zn、Fe、Mn、Mg、Cu的含量;制備血清,測定其白細胞數(shù)及血清IgG含量;脾臟稱重,計算脾臟指數(shù);HE染色觀察大鼠脾組織細胞形態(tài)學變化;采用Trizol提取脾臟中總的RNA,應用熒光定量RT-PCR法測定脾臟中TNF-α、IL-1β和IL-6基因的mRNA相對表達量。[結(jié)果](1)與對照組相比,染鉛組大鼠心、肝、脾、腎、肌肉及血液中鉛的含量明顯升高(P0.05或P0.01);各組織中鈣含量均降低,差異顯著(P0.05);鐵、銅、鋅、錳含量在各組織中均降低,脾中鐵、肝腎中銅、血中鋅含量差異均達顯著或極顯著(P0.05或P0.01);心臟中鎂含量升高,差異極顯著(P0.01)。與染鉛組比較,試驗組大鼠鉛含量降低,試驗Ⅱ組各組織中降低明顯,差異顯著或極顯著(P0.05或P0.01);各組織中鈣含量升高,試驗Ⅰ、Ⅱ組達差異顯著(P0.05);銅、鋅含量在各組織中均有所升高,心、肝、腎、肌肉中的試驗Ⅱ組差異顯著(P0.05)。表明適量的補鈣可以干預鉛致動物機體組織中二價礦物元素的失衡。(2)與對照組比較,染鉛組大鼠脾臟指數(shù)、白細胞總數(shù)、淋巴細胞百分比、血清IgG含量下降(P0.05或P0.01),單核細胞百分比和嗜中細胞百分比升高(P0.05或P0.01)。與染鉛組比較,試驗Ⅱ組的脾臟指數(shù)、白細胞數(shù)及試驗Ⅲ組的血清IgG含量上升,差異均達顯著水平(P0.05);單核細胞百分比和嗜中細胞百分比下降,但差異不顯著。說明補充適量的鈣可以通過調(diào)節(jié)脾臟指數(shù)、白細胞數(shù)和血清IgG干預鉛對免疫細胞的損傷。(3)與對照組比較,染鉛組大鼠脾臟淋巴細胞減少,紅白髓交界不明顯,有部分發(fā)生溶解現(xiàn)象,且淋巴細胞排列稀疏,淋巴小結(jié)數(shù)量較少。與染鉛組比較,試驗Ⅱ組大鼠脾臟淋巴細胞增加,紅白髓分界比較明顯,淋巴細胞基本恢復正常,而試驗Ⅲ組淋巴結(jié)有明顯分散現(xiàn)象。表明補鈣可以有效干預染鉛導致脾臟病理性變化,但應控制在一定范圍內(nèi)。(4)與對照組比較,染鉛組大鼠脾臟中的IL-1β、IL-6、TNF-α的相對表達量降低,差異顯著或極顯著(P0.05或P0.01);與染鉛組比較,三個試驗組IL-1β、IL-6、TNF-α的相對表達量均上升,試驗Ⅱ組差異極顯著(P0.01)。[結(jié)論]膳食中補充適量的鈣可干預機體對鉛的吸收,調(diào)節(jié)鉛致大鼠體內(nèi)Ca、Zn、 Fe、Mn、Mg、Cu的代謝失衡,調(diào)控染鉛機體免疫細胞增值能力及脾臟中的IL-1β、IL-6、 TNF-α基因的mRNA相對表達量?梢,補充一定劑量的鈣對鉛致機體免疫損傷有一定的緩解作用,提高染鉛機體免疫能力。
[Abstract]:Objective : To explore the effect of different doses of calcium on the levels of divalent metal elements , white blood cells , serum IgG , spleen index and spleen tissue morphology , and mRNA expression level of immune - related cytokine genes in lead rats . After 60 days of feeding , whole blood , liver , kidney , spleen , heart and muscle were collected , and the contents of Pb , Ca , Zn , Fe , Mn , Mg and Cu were measured .

Morphological changes of spleen cells in rats were observed by HE staining .
The mRNA relative expression of TNF - 偽 , IL - 1尾 and IL - 6 in spleen was determined by fluorescence quantitative RT - PCR . The content of lead in heart , liver , spleen , kidney , muscle and blood increased significantly ( P0.05 or P0.01 ) .
The contents of iron , copper , zinc and manganese decreased in all tissues , and the difference of Zn in spleen , iron , liver and kidney was significant or very significant ( P0.05 or P0.01 ) . Compared with the lead - stained group , the lead content in the experimental group was decreased , and the difference was significant or very significant ( P0.05 or P0.01 ) .
