鳳果花葉病毒高靈敏度血清學(xué)檢測技術(shù)(英文)

目的:建立基于單克隆抗體的檢測番茄等植物中鳳果花葉病毒(Pep MV)的血清學(xué)方法,為Pep MV的田間調(diào)查和診斷及其科學(xué)防控提供快速實用的檢測技術(shù)。創(chuàng)新點:首次制備了抗Pep MV的高度特異和靈敏的單克隆抗體,并利用制備的單抗建立了3種能特異且靈敏地檢測Pep MV的血清學(xué)方法。方法:以差速離心方法提純的Pep MV粒子作為免疫原免疫BALB/c小鼠,通過雜交瘤技術(shù)獲得了能穩(wěn)定傳代并分泌Pep MV單克隆抗體的雜交瘤細(xì)胞株;雜交瘤細(xì)胞注射到小鼠腹腔獲得單克隆抗體腹水,并以制備單抗為核心,根據(jù)血清學(xué)原理建立檢測植物中Pep MV的雙抗夾心酶聯(lián)免疫吸附試驗(DAS-ELISA)、斑點酶聯(lián)免疫吸附試驗(Dot-ELISA)和組織印跡酶聯(lián)免疫吸附試驗(Tissue print-ELISA)三種血清學(xué)檢測方法;利用田間番茄樣品分析建立的血清學(xué)方法檢測Pep MV的有效性。結(jié)論:利用雜交瘤技術(shù)獲得了6株能分泌高度特異靈敏Pep MV單克隆抗體的雜交瘤細(xì)胞株,以分泌的單抗為核心建立了檢測植株中Pep MV的DASELISA、Dot-ELISA和Tissue print-ELISA三種高度靈敏的血清...
