【摘要】:生物體內(nèi)DNA損傷錯配修復(fù)系統(tǒng)是保證生物體內(nèi)細(xì)胞基因組穩(wěn)定和忠實性傳遞一條重要途徑。人類中遺傳性直腸癌研究表明損傷修復(fù)系統(tǒng)中MSH2基因突變會導(dǎo)致組織中細(xì)胞出現(xiàn)增變表型,但不斷增變表型產(chǎn)生的分子機理并不清楚。水稻tree1是植物特有的MSH7基因突變而致的增變表型,表現(xiàn)為水稻植株分蘗增多,株高變矮,生長時期轉(zhuǎn)換推遲等。分子遺傳學(xué)分析發(fā)現(xiàn)tree1增變表型很大一部分與miR156b/c基因高轉(zhuǎn)錄水平緊密相關(guān),而導(dǎo)致miR156b/c基因錯誤高轉(zhuǎn)錄表達(dá)推測可能是表觀遺傳基因OsLHP1突變造成的。
[Abstract]:In vivo DNA damage mismatch repair system is an important way to ensure the stability and faithfulness of cell genome. Studies on human hereditary rectal cancer show that mutations of MSH2 gene in the damage repair system may lead to phenotypic changes in tissues, but the molecular mechanism of increasing phenotypes is not clear. Rice tree1 is an increasing phenotype caused by plant specific MSH7 gene mutation, which is characterized by more tillers, shorter plant height and delayed growth period. Molecular genetic analysis showed that a large part of the phenotype of tree1 was closely related to the high transcriptional level of miR156b/c gene, and the mis-expression of miR156b/c gene might be caused by the OsLHP1 mutation of epigenetic gene.
