【摘要】:目的:我們擬尋找與無功能型垂體腺瘤的侵襲性相關的重要蛋白,為全面、系統(tǒng)研究無功能型垂體腺瘤侵襲生長的機制提供理論依據,為今后利用蛋白質組學方法全面、系統(tǒng)研究無功能型垂體腺瘤的病因,以及分子靶向治療、基因治療提供新的突破點。方法:收集2015年10月至2016年10月期間新疆維吾爾自治區(qū)人民醫(yī)院神經外科收治的經本院手術治療的18例無功能型垂體瘤病人的腫瘤標本(侵襲性生長9例,非侵襲性生長9例)。用二維凝膠電泳結合質譜技術尋找在無功能型垂體腺瘤中表達的差異性蛋白;應用雙向凝膠電泳(2-DE)結合基質輔助激光解析電離飛行時間質譜技術(MALDI-TOF-MS)對侵襲性無功能型垂體腺瘤及非侵襲性無功能型垂體腺瘤腫瘤組織進行蛋白質翻譯水平的篩選,并比較組織蛋白的異同,尋找與無功能型垂體腺瘤侵襲性相關的蛋白質及可能在臨床診斷中應用的蛋白質。結果:(1)通過雙向電泳結果(通過PDQuest軟件顯不同組的差異表達蛋白質點),建立了無功能型垂體腺瘤的的2-DE圖譜。(2)通過研究我們找到4種差異蛋白點,其中p14ARF蛋白在侵襲性無功能型垂體腺瘤的腫瘤組織中較非侵襲性無功能型垂體腺瘤的腫瘤表達下調,EGFR、HMGA2、PIK3CA蛋白在侵襲性無功能型垂體腺瘤的腫瘤組織中較非侵襲性無功能型垂體腺瘤的腫瘤表達上調。結論:這4種差異蛋白可能與無功能型垂體腺瘤的侵襲性生長有關。
[Abstract]:Objective: to search for important proteins related to invasiveness of non-functional pituitary adenomas, and to provide theoretical basis for a comprehensive and systematic study of the mechanism of invasiveness and growth of non-functional pituitary adenomas. In order to study the etiology and molecular targeted therapy of non-functional pituitary adenoma systematically by proteomics. Gene therapy provides a new breakthrough. Methods:. From October 2015 to October 2016, 18 patients with disfunctional pituitary adenoma treated in neurosurgery department of Xinjiang Uygur Autonomous region people's Hospital were collected (. Invasive growth was found in 9 cases. Two dimensional gel electrophoresis and mass spectrometry were used to search for differentially expressed proteins in non-functional pituitary adenomas. Two-dimensional gel electrophoresis (2-DE) combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MSM). The protein translation level of invasive and non-invasive pituitary adenomas was screened. The differences and similarities of histone were compared. Search for proteins associated with invasiveness of nonfunctional pituitary adenomas and proteins that may be used in clinical diagnosis. PDQuest software was used to show the differentially expressed protein spots in different groups. The 2-DE map of non-functional pituitary adenoma was established. The expression of p14ARF protein was down-regulated in tumor tissues of invasive non-functional pituitary adenomas than that of non-invasive pituitary adenomas. The expression of PIK3CA protein was up-regulated in invasive non-functional pituitary adenomas than in non-invasive pituitary adenomas. Conclusion:. These four differentially expressed proteins may be related to the invasive growth of nonfunctional pituitary adenomas.
