高個體識別力通用單核苷酸多態(tài)性位點(diǎn)篩選及檢測

【摘要】:目的:在全基因組中查找能夠在世界各大人群中通用的高個體識別力單核苷酸多態(tài)性(SNPs)位點(diǎn),以期為SNPs應(yīng)用于法醫(yī)學(xué)個體識別奠定基礎(chǔ)。方法:從國際人類基因組單體型圖計(jì)劃(HapMap)數(shù)據(jù)庫下載270名個體各約4000000個全基因組SNPs分型數(shù)據(jù),運(yùn)用自主編寫的計(jì)算機(jī)程序,按照較小等位基因頻率(MAF)≥0.45的原則提取在中國北京漢族人群、日本東京人群、尼日利亞約魯巴人群以及美國白人人群中同時存在的SNPs,以焦磷酸測序技術(shù)對其中的rs4607417和rs749305在河南漢族人群(n=144)中進(jìn)行群體遺傳學(xué)調(diào)查,計(jì)算Hardy-Weinberg平衡概率(HWP)、雜合度(Het)、多態(tài)信息含量(PIC)和個體識別力(DP),并與HapMap的數(shù)據(jù)(n=45)進(jìn)行比較。結(jié)果:共找到1439個通用的SNPs。在河南漢族人群中,rs4607417和rs749305的HWP分別為0.872和0.423,Het分別為0.476和0.524,PIC分別為0.384和0.388,DP分別為0.616和0.612。與4個人群相比,僅在rs4607417上其等位基因頻率與日本東京人群差異有統(tǒng)計(jì)學(xué)意義(χ2=5.068,P=0.024)。結(jié)論:rs4607417和rs749305是通用的高個體識別力SNPs;前述篩選獲得的1439個SNPs具有高度可靠性和可信度,可用作在世界各人群中通用的個體識別核心SNPs。
