線粒體基因復(fù)合擴(kuò)增進(jìn)行種屬鑒定的研究

【摘要】:目的 生物性檢材的種屬鑒定是法醫(yī)物證學(xué)主要研究內(nèi)容之一。本課題針對(duì)線粒體DNA(Mitochondrial DNA,mtDNA)編碼序列的保守區(qū)和可變區(qū)在人與哺乳動(dòng)物間的異同,設(shè)計(jì)并建立穩(wěn)定可靠的種屬鑒定體系,探索一種適合法醫(yī)學(xué)實(shí)踐的,快速、靈敏、簡便而實(shí)用的種屬鑒別方法,以解決當(dāng)前種屬鑒定中存在的不足。 方法 針對(duì)本課題的研究目的,對(duì)mtDNA的37個(gè)基因進(jìn)行研究,制定目的基因篩選原則,遵循該原則篩選出合適的編碼基因,根據(jù)建立復(fù)合擴(kuò)增體系的要求,,建立穩(wěn)定可靠的線粒體基因復(fù)合擴(kuò)增反應(yīng)體系。復(fù)合擴(kuò)增產(chǎn)物用非變性聚丙烯酰胺凝膠垂直電泳(PAGE)、硝酸銀染色技術(shù)進(jìn)行種屬鑒定。為驗(yàn)證該系統(tǒng)的穩(wěn)定性以及在法醫(yī)學(xué)實(shí)踐中的可行性,進(jìn)行了包括系統(tǒng)靈敏度檢測(cè)、不同環(huán)境下樣本的檢測(cè)、形成于不同基質(zhì)上樣本的檢測(cè)和陳舊樣本等的檢測(cè),從而對(duì)其法醫(yī)學(xué)應(yīng)用價(jià)值進(jìn)行評(píng)估。
[Abstract]:Objective to identify the species of biologic samples is one of the main contents of forensic forensics. This paper aims at the similarities and differences of the conserved and variable regions of mitochondrial DNA(Mitochondrial mtDNAs between human and mammal. To design and establish a stable and reliable species identification system, and to explore a rapid, sensitive, simple and practical method suitable for forensic practice, in order to solve the shortcomings of current species identification. Methods for the purpose of this study, 37 genes of mtDNA were studied, and the screening principles of target genes were worked out. According to the requirements of establishing a multiplex amplification system, appropriate coding genes were screened according to the principle. A stable and reliable multiplex amplification system of mitochondrial gene was established. The products were identified by polyacrylamide gel electrophoresis and silver nitrate staining. Feasibility in the practice of forensic medicine, It includes system sensitivity detection, sample detection in different environments, sample detection on different matrix and old sample detection, so as to evaluate the value of forensic application.
