【摘要】:為探討N-乙酰半胱氨酸(NAC)對(duì)膿毒血癥大鼠腸損傷的影響及對(duì)炎癥因子表達(dá)的影響,選取清潔級(jí)SD大鼠30只,體重250~300g,隨機(jī)分為假手術(shù)組(Sham組)、膿毒癥組(SEP組)和N-乙酰半胱氨酸組(NAC組),每組10只;Sham組僅開(kāi)腹,暴露盲腸,NAC組和SEP組行盲腸結(jié)扎穿孔術(shù),再分別靜脈給予150mg·kg~(-1)NAC和等量生理鹽水;3組均于再灌注4h處死大鼠,觀察小腸組織病理學(xué)結(jié)果;ELISA檢測(cè)血中腸脂肪酸結(jié)合蛋白(I-FABP)和D-乳酸濃度;ELISA法和Western blot法檢測(cè)血清和小腸中TNF-α、IL-6、IL-10濃度。結(jié)果表明:光鏡下Sham組未出現(xiàn)病理?yè)p傷,SEP組損傷重,NAC組損傷減輕;與Sham組比較,SEP組I-FABP、D-乳酸顯著升高(p0.05);與SEP組比較,NAC組I-FABP、D-乳酸顯著降低(p0.05);與Sham組比較,SEP組血清中TNF-α、IL-6、IL-10顯著升高(p0.05);與SEP組比較,NAC組TNF-α、IL-6顯著降低(p0.05),IL-10顯著升高(p0.05),小腸炎癥相關(guān)因子變化趨勢(shì)同血清檢測(cè)。N-乙酰半胱氨酸對(duì)膿毒血癥大鼠腸黏膜屏障有保護(hù)作用,其機(jī)制與抑制炎癥反應(yīng)有關(guān)。
[Abstract]:To investigate the effects of N-acetylcysteine (NACN) on intestinal injury and the expression of inflammatory cytokines in sepsis rats, 30 SD rats of clean grade were selected, weighing 250 ~ 300g. They were randomly divided into sham-sham group, septic group (SEP group) and N-acetylcysteine group (NAC group, n = 10 in each group). In the Sham group, the caecum ligation and perforation were performed in the exposed caecum group and the SEP group, and were given 150 mg 路kg~(-1)NAC and the same amount of normal saline respectively. All the rats in the 3 groups were killed at 4 h after reperfusion, and the histopathological results of small intestine were observed. The concentrations of serum fatty acid binding protein (I-FABP) and D-lactic acid were detected by ELISA. ELISA and Western blot were used to detect the concentration of IL-10 in serum and small intestine. The results showed that there was no pathological injury in Sham group under light microscope. The damage of SEP group was alleviated. Compared with Sham group, I-FABPnD-lactic acid in SEP group was significantly higher than that in Sham group (P 0.05). Compared with SEP group, I-FABP- D- lactic acid in NAC group was significantly lower than that in SEP group. Compared with the Sham group, the serum TNF- 偽 IL-6 + IL-10 in the Sham group was significantly higher than that in the Sham group (P 0.05). Compared with SEP group, TNF- 偽 IL-6 in NAC group decreased significantly (P 0.05) and IL-10 increased significantly (p 0.05). The change of inflammatory related factors in small intestine is related to the protective effect of serum. N-acetylcysteine on intestinal mucosal barrier in rats with sepsis, and its mechanism is related to the inhibition of inflammatory reaction.
