蒙藥森登-4復(fù)方及其單味藥的多糖成分提取及抗氧化作用研究

【摘要】:多糖是蒙藥森登-4及其各單味藥的重要活性成分之一。具有增強人體免疫、降血脂、降血糖、抗腫瘤、抗細菌、抗病毒、抗輻射、抗凝血、抗衰老、抗氧化等作用。本論文首先對森登-4及其各單味藥中多糖的提取工藝進行研究,試驗中以多糖含量為指標,以固液比、提取時間、提取溫度和提取次數(shù)為因素,使用響應(yīng)面設(shè)計優(yōu)化了蒙藥森登-4復(fù)方及其單味藥的多糖最佳水提工藝;其次,探討了蒙藥森登-4復(fù)方及其各單味藥中多糖對羥基自由基(·OH-)、超氧陰離子自由基(O2-)、亞硝酸根離子(NO2-)和DPPH自由基的清除能力以及多糖的總還原能力,從中探討蒙藥森登-4配伍前后抗氧化活性的變化。多糖提取工藝的研究結(jié)果表明,森登-4及其單味藥的最佳提取條件各不相同。(1)森登-4多糖的最佳提取工藝為:固液比1:40,提取溫度為95℃,提取時間為3 h,提取次數(shù)為3次,多糖提取百分含量為8.90%。(2)文冠木多糖最佳提取工藝為:固液比1:32,提取溫度為69℃,提取時間為1.8 h,提取次數(shù)為2次,多糖提取百分含量為1.45%。(3)川楝子多糖的最佳提取工藝為:固液比1:20,提取溫度為90℃,提取時間為2.9 h,提取次數(shù)為3次,多糖提取百分含量為22.34%。(4)梔子多糖的最佳提取工藝為:固液比1:33.7,提取溫度為80℃,提取時間為3 h,提取次數(shù)為3次,多糖提取百分含量為10.97%。(5)訶子多糖的最佳提取工藝為:固液比1:28,提取溫度為69℃,提取時間為1 h,提取次數(shù)為3次,多糖提取百分含量為5.99%。多糖抗氧化研究表明,配伍前后藥材多糖的抗氧化活性發(fā)生變化,且森登-4多糖的抗氧化活性優(yōu)于其四種單味藥。(1)森登-4多糖清除·OH-的能力強于對照品Vc、文冠木、梔子、川楝子,弱于訶子。其中梔子清除羥基自由基的能力達26.35%。(2)森登-4多糖清除O2-的能力強于對照品甘露醇和其他四種藥材。(3)森登-4多糖清除NO2-的能力強于Vc、訶子、川楝子,弱于梔子,與文冠木相當。(4)川楝子和訶子清除DPPH的能力弱于Vc。森登-4、文冠木和梔子多糖對DPPH的清除作用未能測試到。(5)蒙藥森登-4及其各單味藥中多糖的還原能力弱于對照品Vc的還原能力,且森登-4的還原能力也弱于其他四中單味藥。
[Abstract]:Polysaccharide is one of the important active components of Mongolian medicine Sendeng-4 and its single drugs. It has the functions of enhancing human immunity, lowering blood lipid, lowering blood glucose, anti-tumor, anti-bacterial, anti-virus, anti-radiation, anticoagulant, anti-aging, antioxidant and so on. In this paper, the extraction technology of polysaccharide from Sendeng-4 and its single drug was studied. In the experiment, the optimal water extraction process of polysaccharide from Sendeng-4 compound and its single drug was optimized by using response surface design, taking the content of polysaccharide as the index, the ratio of solid to liquid, extraction time, extraction temperature and extraction times as factors. Secondly, the scavenging ability of polysaccharides to hydroxyl radical (OH -), superoxide anion radical (O2 -), nitrous acid ion (NO2-) and DPPH radical and the total reduction ability of polysaccharide in Mongolian medicine Sendeng-4 compound and its single drugs were discussed, and the changes of antioxidant activity before and after the compatibility of Mongolian medicine Sendeng-4 were discussed. The results showed that the optimum extraction conditions of Sendeng-4 and its single drug were different.
